Neural Disease Mechanisms Across Scales: Insights from Advanced 2D and 3D Models
This Research Topic examines how integrated neural models can reveal previously unrecognized mechanisms of neurological disorders across biological scales, linking genetic and molecular dysregulation to cellular, synaptic, network, and circuit level dysfunction. A central aim is to better understand how disease mechanisms emerge across levels of organization and how these insights may support new therapeutic strategies. We particularly welcome studies using advanced 3D neural models, including:• Biofabricated and bioprinted tissues• iPSC-derived neuronal and glial models• Engineered and fabricated neural platforms• Microfabricated systems• Organ-on-chip approaches• Assembloids and organoid-based systemsWe also welcome complementary studies using advanced 2D systems when these provide strong mechanistic insight, enable validation of disease-relevant phenotypes, or are combined with more complex 3D approaches.Studies of particular interest include those that:• Reveal novel disease mechanisms• Establish causal links between genotype and neural phenotype• Identify synaptic and circuit dysfunction as drivers of disease onset or progression• Integrate electrophysiology, imaging, omics, gene editing, or systems-level analysis• Provide mechanistic insight with relevance for therapeutic interventionThis Research Topic is cross-listed with Frontiers in Materials (Biomaterials and Bio-Inspired Materials) to bring in expertise from the biomaterials community. Manuscripts centred on scaffold design, bio-inks, biomaterial characterisation or microfabrication strategies for neural applications are best suited to that sister section, while studies whose main contribution is the molecular or circuit-level neuroscience should remain with Frontiers in Molecular Neuroscience.
