Neuro-Tumor Crosstalk: Mechanisms and Therapeutic Opportunities in Cancer Neuroscience
The emerging field of Cancer Neuroscience has revealed that the nervous system plays a pivotal role in cancer initiation, progression, and metastasis. Tumors are not merely innervated; they actively recruit nerve fibers through neoneurogenesis and exploit neurotransmitters and neuropeptides as signaling molecules to promote their growth. Conversely, tumors can remodel neural networks and influence systemic neuronal function. This bidirectional communication highlights nerves as an integral component of the tumor stroma and a promising target for therapeutic intervention. This Research Topic aims to explore the multi-dimensional landscape of neuro-tumor interactions, spanning from fundamental mechanistic studies to clinical translation. By integrating insights from multidisciplinary research, we seek to elucidate how the nervous system shapes tumor progression, with a particular focus on immune responses, the tumor microenvironment (TME), and adaptive mechanisms of cancer cells. Our goal is to identify novel therapeutic targets and strategies, accelerating the translation of basic findings into innovative treatments that specifically modulate neuro-tumor crosstalk. We welcome Original Research, Reviews, and Mini-Reviews covering all aspects of neuro-tumor interactions. Specific topics of interest include, but are not limited to:• Molecular mechanisms of perineural invasion (PNI) and its clinical relevance.• Roles of neurotransmitters and neuropeptides in tumor proliferation, migration, and metastasis.• Influence of chronic stress and sympathetic/parasympathetic nervous system activity on cancer progression.• Interactions between the nervous system and the tumor immune microenvironment.• Pre-clinical and clinical studies on repurposing neurological drugs for cancer therapy.• Denervation strategies and their therapeutic outcomes.• Studies on both solid tumors and hematological malignancies, provided they offer novel insights into neural regulation.By bringing together research across disciplines, this Research Topic aims to advance our understanding of neuro-tumor crosstalk and foster the development of innovative therapeutic strategies targeting this emerging aspect of cancer biology.
