New Approach Methodologies in Immunotoxicology
Immunotoxicology has historically relied on in vivo animal models to assess the potential adverse effects of chemicals, pharmaceuticals, and environmental agents on the immune system. However, these traditional approaches often face limitations regarding predictive accuracy for human outcomes, cost, and throughput. The rapid evolution of New Approach Methodologies (NAMs) represents a transformative shift toward human-relevant, mechanism-based safety assessments. By integrating advanced alternative tools, researchers can better understand complex immunological pathways and improve the predictability of chemical-induced immunotoxicity. This Research Topic aims to collect the latest advancements in the development, validation, and application of NAMs specifically tailored for immunotoxicology. We seek to bring together innovative research that addresses key challenges in translating these alternative methods into regulatory frameworks, establishing robust validation guidelines, and mapping complex immune responses. By bridging the gap between innovative methodology and practical risk assessment, this collection will help shape the future of chemical safety evaluation. We welcome submissions of original research, reviews, mini-reviews, and perspective articles focusing on, but not limited to, the following areas:- High-throughput screening assays for immunotoxic hazard identification- In vitro and 3D organotypic models of the human immune system, including organ-on-a-chip platforms- Computational toxicology, quantitative structure-activity relationship (QSAR) models, and machine-learning approaches- Adverse Outcome Pathways (AOPs) related to immunosuppression, sensitization, and autoimmunity- Biomarkers of immunotoxicity identified through multi-omics technologies- Case studies and regulatory perspectives demonstrating the integration of NAMs into risk assessment workflows.
