New avenues for the development of advanced immunotherapies: capitalizing on studies of the B and T cell receptor repertoire - Volume II

The adaptive immune receptor repertoire represents a dynamic and highly diverse system that underpins immune protection, immune-mediated pathology, and therapeutic response. Advances in high-throughput sequencing technologies, single-cell multi-omics, and computational analytics have transformed our capacity to characterize T cell receptor (TCR) and B cell receptor (BCR) repertoires at unprecedented resolution. This second volume (visit Vol.I here) aims to consolidate recent progress presented at the 1st Immunorepertoire and Biologics Development conference (IRCCS IRST, Meldola) dedicated to immunorepertoires and to provide an updated platform for cutting-edge research in the field. The goal of this Research Topic is to highlight methodological innovations, analytical frameworks, and translational applications of immune repertoire profiling. By bringing together technological, computational, and clinical perspectives, this collection seeks to foster integration across disciplines and to accelerate the implementation of immunorepertoire-based approaches in immunotherapy, diagnostics, and immune monitoring.Over the past decade, adaptive immune receptor repertoire sequencing (AIRR-seq) has evolved from a descriptive tool into a quantitative and mechanistic approach for studying immune responses. Improvements in library preparation, single-cell sequencing, and bioinformatics pipelines now allow precise mapping of clonal expansion, somatic hypermutation, antigen specificity, and lineage relationships. These advances have profoundly impacted research in cancer immunotherapy, infectious diseases, autoimmunity, vaccination, and transplantation. At the same time, challenges remain in data standardization, repertoire annotation, cross-study comparability, and functional interpretation. The growing integration of repertoire data with transcriptomics, epigenomics, and spatial technologies is reshaping our understanding of adaptive immunity and opening new translational opportunities. This Research Topic welcomes contributions arising from the 1st Immunorepertoire and Biologics Development conference (IRCCS IRST, Meldola) as well as related submissions addressing innovative aspects of TCR and BCR repertoire research. Topics of interest include, but are not limited to:● Technological advances in bulk and single-cell TCR/BCR sequencing● Computational methods and machine learning approaches for repertoire analysis● Innovative screening methodologies involving NGS and complex libraries of biologics to derive novel therapeutics for human diseases● Clonal dynamics and immune monitoring in cancer, infection, autoimmunity, and vaccination● Antigen receptor diversity, convergence, and public clonotypes● Functional interpretation of immune repertoires● Standardization, reproducibility, and data-sharing initiatives● Integration of immunorepertoire data with multi-omics and spatial platforms● Translational applications in immunotherapy and precision medicineWe welcome manuscripts that advance conceptual, technical, or translational understanding of adaptive immune receptor repertoires.
