Next-Generation Nanomedicine for Therapy-Resistant Solid Tumors
Therapeutic resistance remains a major challenge in the treatment of solid tumors, significantly limiting the long-term efficacy of conventional chemotherapy, targeted therapies, and immunotherapies. Resistance arises through diverse molecular and cellular mechanisms, including oncogenic signaling rewiring, cancer cell plasticity, metabolic adaptation, immune evasion, cancer stem cell maintenance, and tumor microenvironment–mediated protection. Recent advances in nanomedicine have enabled the development of targeted delivery systems capable of modulating these mechanisms and enhancing therapeutic efficacy. This Research Topic aims to highlight emerging nanomedicine-based strategies designed to investigate, target, and overcome the molecular and cellular determinants of therapeutic resistance in solid tumors. Areas of interest include polymeric and lipid-based nanocarriers, stimuli-responsive systems, and combination therapy approaches that address dysregulated signaling pathways, tumor metabolism, cancer stemness, immune resistance, and tumor microenvironment interactions. Particular emphasis will be placed on studies providing mechanistic insight into therapeutic response and resistance, supported by experimental validation in relevant cellular, organoid, or in vivo models.By bringing together contributions from oncology, molecular biology, cancer immunology, and nanomedicine, this Topic seeks to advance the mechanistic understanding of therapy resistance and support the development of next-generation nanotherapeutic strategies for improving outcomes in resistant cancers. Please note: manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this Research Topic.
