Next-Generation Nanoscale Delivery Solutions for Nucleic Acid Therapies
Nanotechnology is playing a pivotal role in revolutionizing the delivery of nucleic acid therapeutics. Through the development of precisely engineered nanomaterials, such as lipid nanoparticles, polymers, dendrimers, and hybrid nanosystems, researchers have achieved significant improvements in the stability, targeted delivery, and efficiency of transporting nucleic acids into cells. Breakthroughs in the use of nanotechnology-facilitated mRNA vaccines have showcased the remarkable potential of these platforms to overcome barriers that have historically hindered the clinical translation of therapies based on DNA, RNA, and gene-editing tools. However, despite these advances, key challenges persist, including issues of targeted delivery, endosomal escape, scalable manufacturing, and immunogenicity, which continue to limit the full potential of nucleic acid-based treatments. This Research Topic aims to advance the field of nanotechnology by fostering new strategies, insights, and collaborative efforts focused on the development of safe and effective multicomponent delivery systems for nucleic acid therapeutics. The primary goal is to gather innovative research that addresses outstanding obstacles in the design, optimization, and application of nanotechnology-based carriers, facilitating more precise and efficient gene modulation and therapy. By highlighting cutting-edge discoveries and emerging concepts, this Research Topic seeks to pave the way for novel approaches and future advancements in the field. To gather further insights within the nanotechnology landscape of multicomponent nucleic acid delivery platforms, we welcome articles addressing, but not limited to, the following themes:• Engineering and optimization of nanoscale carriers for nucleic acid delivery• Advances in lipid, polymeric, and hybrid nanoparticles for therapeutic applications• Mechanisms of cellular uptake and endosomal escape enabled by nanotechnology• Targeted and controlled release strategies for nucleic acids using nanomaterials• Minimizing immunogenicity and toxicity of nanoscale delivery systems• Overcoming biological barriers and improving in vivo efficacy with nanotechnology• Translational and manufacturing challenges for nanotechnology-based nucleic acid therapiesWe invite original research, reviews, perspectives, and methodological papers that advance the understanding and practical development of nanotechnology-driven multicomponent nucleic acid delivery systems.Topic Editor Hasan Uludag is a shareholder in RJH Biosciences.
