Novel Approaches to Drug Mechanism and Discovery in Hemostasis and Thrombosis

Thrombosis and hemostasis are vital processes preserving vascular health, with disruptions leading to debilitating and life-threatening disorders such as hemophilia, von Willebrand disease, thrombotic microangiopathies (TMAs), thrombophilias, and immune thrombocytopenia. With the advent of advanced molecular and pharmacological techniques, there has been unprecedented progress in our understanding of the drug mechanisms underpinning these disorders and the development of innovative therapies. This Research Topic is dedicated to addressing critical gaps in experimental pharmacology and drug discovery for bleeding and thrombotic disorders—spanning rare congenital deficiencies to acquired immunological conditions. We focus on cutting-edge studies that elucidate the molecular, cellular, and pharmacodynamic mechanisms of existing and investigational drugs, as well as innovative strategies to identify, validate, and develop new therapeutic agents. Areas of keen interest include the preclinical and early clinical characterization of small molecules, biologics, gene therapies, targeted inhibitors (e.g., complement and immune modulators), non-factor replacement therapies, and next-generation anticoagulants. We welcome original research, review articles, clinical trials, and general commentary that explore the following areas, including but not limited to:• Identification and validation of novel drug targets involved in coagulation, platelet biology, and immune-mediated mechanisms.• High-throughput screening, in-vitro and in-vivo models, and translational studies that accelerate drug discovery and early pipeline development.• Characterization of drug-receptor or target interactions, signaling pathways, and resistance mechanisms.• Preclinical and first-in-human data on new agents, including efficacy, safety, and pharmacokinetic/pharmacodynamic assessments.• Use of big data, artificial intelligence, and precision medicine to identify biomarkers predictive of drug response or adverse effects.• Studies and elucidation of unexpected drug effects or mechanisms in the context of hemostatic and thrombotic disorders.
