Novel Potential Approaches to Overcome Immunotherapy Resistance in Solid Tumors
The field of oncology has witnessed significant advancements with the introduction of novel immunotherapies, including immune checkpoint inhibitors, chimeric antigen receptor T (CAR-T) cells, vaccines, and oncolytic viruses. These innovative therapies have transformed the management of various malignancies, offering improved survival outcomes for many cancer patients compared to conventional treatment modalities. However, a substantial subset of patients experience only transient benefits, with some showing rapid disease progression. This phenomenon is primarily driven by the occurrence of acquired and innate resistance to immunotherapy. Research has identified multiple tumor-intrinsic and tumor-extrinsic mechanisms that contribute to this resistance, underscoring the need for a deeper understanding of these pathways. This Research Topic aims to delve into the pivotal mechanisms underlying resistance to immunotherapy and explore promising therapeutic strategies to overcome these challenges. By integrating insights from multi-omics approaches, biomarker discovery, and innovative combinatorial treatment regimens, we aim to enrich the current understanding and identify new avenues for clinical intervention. Our objective is to foster a multidisciplinary perspective, bridging basic research with clinical application, to translate emerging concepts into actionable strategies that can enhance patient outcomes. Emphasizing the potential of combining therapies and leveraging biomolecular insights, this Research Topic seeks to address the ongoing battle against immunotherapy resistance in solid tumors. To gather further insights in the quest to tackle immunotherapy resistance in solid tumors, we welcome articles addressing, but not limited to, the following themes:- Mechanisms of innate and acquired resistance to immunotherapy.- Tumor-intrinsic factors influencing resistance pathways.- Tumor-extrinsic factors, including the tumor microenvironment and immune components.- Multi-omics approaches to identify biomarkers of resistance.- Innovative combinatorial regimens to counter resistance.- Translational strategies for the clinical application of novel findings.- Case studies highlighting resistance patterns and breakthrough therapies. Please note that manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases that are not accompanied by validation (independent cohort or biological validation in vitro or in vivo) are out of the scope of this Research Topic.Dr. Kurzrock has received past or current research funding from Boehringer Ingelheim, Debiopharm, Foundation Medicine, Genentech, Grifols, Guardant, Incyte, Konica Minolta, Medimmune, Merck Serono, Omniseq, Pfizer, Sequenom, Sysmex, Takeda, and TopAlliance and from the NCI; as well as consultant and/or speaker fees and/or advisory board/consultant for Actuate Therapeutics, AstraZeneca, Bicara Therapeutics, Inc., Biological Dynamics, Boehringer Ingelheim, Caris, Daiichi, Datar Cancer Genetics, EISAI, EMD Serono, EOM Pharmaceuticals, Iylon, Jackson Laboratories, LabCorp, Lanauria Therapeutics, Merck, NeoGenomics, Neomed, Pfizer, Precirix, Prosperdtx, Quanta Therapeutics, Recordati, Regeneron, Roche, Ryght AI, TD2/Volastra, Tempus, Turning Point Therapeutics, X-Biotech; has an equity interest in CureMatch Inc.; serves on the Board of CureMatch and CureMetrix and XZOM, and is a co-founder of CureMatch.
