Novel therapeutic targets and emerging treatment strategies in vasculitides

Vasculitides encompass a wide spectrum of diseases affecting vessels of various sizes, including large-, medium-, and small-vessel forms. Advances in immunology and molecular biology have provided new insights into disease pathogenesis, including the roles of immune dysregulation, autoantibodies, cytokine networks, endothelial injury, and genetic susceptibility. These discoveries have already led to the development of targeted biologic therapies that have changed clinical practice in several vasculitis conditions. However, many aspects of disease mechanisms remain incompletely understood, and treatment responses vary significantly among patients. There is an urgent need to identify additional therapeutic targets and explore innovative treatment approaches that are both effective and safer. Ongoing research across multiple disciplines continues to reveal new pathways and strategies that may transform the management of vasculitis in the coming years.Vasculitides comprise a heterogeneous group of inflammatory disorders characterized by immune-mediated damage to blood vessels of different sizes and organs. Although significant advances have been made in disease classification and management, many patients continue to experience refractory disease, treatment-related toxicity, or relapses. Current therapies often rely on broad immunosuppression, underscoring the need for more targeted and personalized approaches. This Research Topic aims to bring together cutting-edge research focused on identifying novel therapeutic targets and developing emerging treatment strategies across all types of vasculitis. By integrating basic, translational, and clinical research, we seek to advance understanding of the molecular and immunological mechanisms that drive vascular inflammation and tissue injury. The ultimate goal is to promote the discovery of innovative interventions that improve disease control, reduce long-term complications, and support precision medicine approaches for patients with vasculitides. This Research Topic welcomes contributions addressing:- Novel therapeutic targets and emerging treatment strategies across the full spectrum of vasculitides, including but not limited to ANCA-associated vasculitis, large-vessel vasculitis, immune complex–mediated vasculitis, and organ-specific forms. - Mechanistic insights into vascular inflammation- Identification of molecular targets, development of biologics or small-molecule therapies, immune modulation strategies, biomarker discovery for treatment response, and approaches to personalized medicine- Clinical trials, translational research, experimental models, and real-world evidence evaluating new or repurposed therapies are particularly encouraged. We welcome a variety of manuscript types, including Original Research Articles, Reviews, Mini-Reviews, Clinical Studies, and Perspective articles. Contributions that bridge basic science and clinical application, and those proposing future therapeutic directions, are especially encouraged.
