Osteoimmunology: Advancing Understanding and Therapies Through Immune-Bone Interactions in Autoimmune, Inflammatory, and Autoinflammatory Diseases

Osteoimmunology, an interdisciplinary field bridging bone biology and immunology, has significantly advanced our understanding of the reciprocal communication between the skeletal and immune systems. Over the past two decades, research in this area has revealed that immune dysregulation can profoundly impact bone metabolism, particularly in autoimmune diseases like rheumatoid arthritis, leading to transformative developments in both diagnostics and therapeutics. This interplay extends beyond classic rheumatological disorders, influencing outcomes in metabolic bone diseases such as osteoporosis, oncologic conditions involving bone metastases, as well as hematological malignancies like acute myeloid leukemia and various histiocytosis. More recent evidence suggests that bone-immune interactions are also pivotal in conditions traditionally managed within endocrinology and gastroenterology, including type 1 diabetes mellitus, autoimmune thyroiditis, celiac disease, and chronic inflammatory bowel diseases. While the shared molecular and cellular pathways have become increasingly apparent, significant gaps remain, especially concerning the mechanisms leading to bone dysmetabolism in the context of autoinflammatory diseases, an area that is only beginning to be explored. This Research Topic aims to illuminate new pathogenetic pathways and foster a deeper understanding of the mechanisms underpinning immune-mediated bone disorders across a spectrum of clinical settings. By highlighting both established and emerging concepts, we seek to refine current models of bone and immune system crosstalk and explore innovative therapeutic opportunities. Specific goals include elucidating the consequences of immune cell and bone cell senescence, evaluating the effects of existing drugs on bone health in immune-mediated diseases, and identifying shared and disease-specific pathways that could serve as drivers for future treatment innovations. To gather further insights in the interdisciplinary application of osteoimmunology to autoimmune, inflammatory, and autoinflammatory diseases, we welcome articles addressing, but not limited to, the following themes:• Mechanistic studies of bone-immune system crosstalk in autoimmunity, inflammation, and autoinflammation• Pathogenetic insights and novel hypotheses in bone metabolism disorders linked to immune dysregulation• The role of cellular and molecular senescence in immune and bone cell aging• Therapeutic impacts of current and emerging immunomodulatory drugs on bone health• Translational studies bridging experimental findings to clinical applications in rheumatology, endocrinology, gastroenterology, oncology, and hematology• Unexplored or emerging clinical contexts, such as bone involvement in autoinflammatory diseasesWe encourage submission of original research, reviews, case studies, and perspective articles to provide comprehensive coverage of this evolving field.
