Osteoimmunology: Bone, Immune, and Endocrine Crosstalk in Health, Aging, and Disease

Bone is a dynamic organ that engages in complex communication with the immune and endocrine systems. The concept of osteoimmunology has revolutionized our understanding of skeletal physiology and disease by revealing the deep interconnections between bone remodeling, immune responses, and hormonal regulation. Osteoblast lineage cells and osteoclasts actively interact with immune cells directly or through cytokines, growth factors, and hormones to maintain skeletal homeostasis. Moreover, endocrine factors such as parathyroid hormone, vitamin D, sex hormones, glucocorticoids, and adipokines play dual roles in regulating both immune function and skeletal metabolism, underscoring the tri-directional relationship among these systems. In contrast, dysregulation in bone-immune-endocrine crosstalk contributes to the development of bone and metabolic disorders such as osteoporosis, rheumatoid arthritis, diabetes-related bone disease, and cancer-induced bone loss. This special collection aims to advance our understanding of the intricate relationships between the bone, immune, and endocrine systems and highlight the gaps as well as emerging opportunities for translational interventions that promote skeletal and systemic health. We are particularly focused on studies exploring how endocrine and immune systems regulate bone homeostasis and how dysregulation drives skeletal fragility and chronic inflammatory bone conditions. Studies describing innovative therapeutic strategies targeting the bone-immune-endocrine pathways to preserve bone health and metabolism across the lifespan are also encouraged. Therefore, we welcome submissions covering, but are not limited to, the following themes:Molecular and cellular mechanisms involved in bone–immune–endocrine crosstalk• The impact of immune and hormonal signaling on bone remodeling and regeneration• Endocrine regulation of immune cell differentiation and inflammatory bone loss• The role of osteoimmunology in age-related bone and metabolic disorders, including the osteoimmune–brain axis and its potential links to cognitive decline, dementia, and Alzheimer’s disease• Bone–immune interactions in endocrine diseases such as diabetes and thyroid dysfunction• The influence of adipokines, cytokines, and growth factors on skeletal and immune health• The interaction between the gut microbiome and immune system and its impact on hormonal regulation of bone remodeling and its role in bone diseases.• Possible role of the immune system in disuse-induced bone loss.• Novel therapeutic approaches targeting osteoimmunological pathways in endocrine bone disorders Topic editors: Hamid Yousf Dar, Sandeep Kumar Source: https://www.frontiersin.org/research-topics/75908/osteoimmunology-bone-immune-and-endocrine-crosstalk-in-health-aging-and-disease
