Overcoming Resistance and Advancing Therapeutics in Metastatic Skin Cancer

Despite remarkable advances in immunotherapy and targeted agents, therapeutic resistance remains the central obstacle in managing metastatic skin cancer. In both melanoma and non-melanoma subtypes, tumors frequently adapt through mechanisms of plasticity, clonal evolution, and immune escape, limiting the durability of even the most promising treatment regimens. Understanding how resistance emerges, and how it can be anticipated or overcome, is now one of the most urgent priorities in the field.This Research Topic focuses specifically on the biology of resistance and the development of strategies to address it. We welcome submissions including but not limited to:• Molecular and cellular mechanisms of primary and acquired resistance to immunotherapy, targeted agents, or combination regimens in metastatic skin cancers.• Novel therapeutic combinations, including next-generation immune checkpoint blockade, bispecific approaches, and co-targeting strategies, designed to prevent or overcome adaptive escape.• Tumor plasticity, phenotype switching, and immune evasion as drivers of treatment failure in melanoma and cutaneous malignancies.• Preclinical and early-phase clinical data on emerging agents or combination strategies targeting resistance pathways.We particularly encourage submissions that integrate mechanistic insight with therapeutic strategy moving from “why resistance happens” to “how to break it.” We invite original research articles, systematic reviews, clinical trial reports, and perspective pieces.Please note: manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.
