Pharmacological Advances in Metabolic Syndrome: Beyond Glycemic Control
Metabolic syndrome and related cardiometabolic diseases remain significant challenges in modern medicine owing to their rising prevalence and complex pathophysiology. Despite advances in glucose-lowering agents, patients continue to experience elevated risks for cardiovascular, renal, and hepatic complications. In recent years, incretin-based pharmacotherapies—including GLP-1, GIP, and dual/triple agonists—have garnered substantial attention for their potential to impact metabolic regulation beyond glycemic control. Studies have shown that these agents not only improve insulin secretion and glycemic indices but also possess multi-organ benefits, such as cardioprotective, nephroprotective, and hepatoprotective effects. Nevertheless, questions remain regarding the precise molecular and cellular mechanisms driving these benefits, the anti-inflammatory and antioxidative properties of incretin-related drugs, and how these translate into improved clinical outcomes. This Research Topic aims to advance our understanding of the latest pharmacological innovations in the management of metabolic syndrome, with a special focus on incretin-based therapies. We seek contributions that elucidate the biological underpinnings of GLP-1, GIP, and related agonists, as well as translational and clinical studies exploring their efficacy, safety, and long-term impact in real-world settings. The goal is to foster dialogue on unresolved issues—such as mechanisms mediating organ-specific benefits, optimal patient selection, and the integration of novel drugs into personalized treatment regimens. Researchers are encouraged to explore combination therapy strategies, emerging drug platforms, and future perspectives that may shape the next generation of treatments for metabolic and cardiometabolic diseases. This Research Topic is broadly focused on pharmacological advances targeting metabolic syndrome and cardiometabolic diseases, while maintaining specific emphasis on incretin-based therapies and their systemic effects. Submissions may address basic, translational, or clinical aspects relevant to these advances. To gather further insights within these boundaries, we welcome articles addressing, but not limited to, the following themes:Molecular and cellular mechanisms of GLP-1, GIP, and dual/triple agonistsThe role of incretin-based therapies in metabolic syndrome, obesity, and type 2 diabetesCardiovascular, renal, and hepatic effects of GLP-1/GIP agonistsAnti-inflammatory and antioxidative actions of incretin-based pharmacotherapiesPreclinical models of metabolic syndrome and cardiometabolic diseasesTranslational and clinical studies evaluating efficacy, safety, and long-term outcomesCombination therapies and personalized pharmacological approachesEmerging incretin-based drugs and future therapeutic perspectivesRelevant article types for this Research Topic include original research, reviews, mini-reviews, perspectives, clinical trials, case reports, and opinion pieces.
