Post-Traumatic Epileptogenesis: Mechanisms, Biomarkers, and Therapeutic Strategies
Traumatic brain injury (TBI) is a major cause of acquired epilepsy, particularly in young adults and military personnel. Epileptogenesis, the process by which an initially non-epileptic brain becomes capable of generating recurrent spontaneous seizures, remains incompletely understood. Post-traumatic epileptogenesis (PTE) involves a dynamic interplay of molecular, cellular, and circuit-level changes, including synaptic reorganization, neuroinflammation, blood-brain barrier disruption, and maladaptive plasticity of inhibitory and excitatory networks. This Research Topic aims to bring together cutting-edge studies that illuminate the mechanisms underlying the transformation from brain injury to chronic epilepsy. By fostering an interdisciplinary discussion, this Research Topic aims to accelerate the development of diagnostic tools and disease-modifying treatments for PTE, ultimately reducing the burden of epilepsy after neurological trauma. Contributions from researchers in basic neuroscience, translational models, and clinical studies are highly encouraged. In particular, we welcome all article types accepted by the journal (including original research and reviews) that address, but are not limited to:• The structural and functional alterations of inhibitory interneurons and excitatory circuits after brain trauma• Neuroinflammatory and glial responses contributing to PTE• The role of extracellular matrix components and perineuronal nets in circuit remodeling• Calcium signaling dysregulation and ion channel pathophysiology following trauma• Pharmacological and molecular interventions aimed at preventing or modifying epileptogenesis• Advanced imaging or electrophysiological biomarkers for early detection and monitoring of epileptogenic progression• The impact of genetic or environmental modifiers on individual risk of developing PTE• Translational models that bridge basic mechanistic insights to human TBI-related epilepsy Topic editors: Feng Gu, Haifeng Shu, Xiaoming Jin Source: https://www.frontiersin.org/research-topics/75634/post-traumatic-epileptogenesis-mechanisms-biomarkers-and-therapeutic-strategies
