PPAR Signaling in Glial Cells: Mechanisms and Therapeutic Insights
Peroxisome proliferator-activated receptor (PPAR) signaling is reshaping our understanding of glial cell biology, especially within the context of demyelinating and inflammatory diseases. Recent advances underscore the importance of distinct PPAR isoforms (PPAR-α, PPAR-β/δ, and PPAR-γ) in the regulation of non-neuronal cells such as astrocytes, microglia, and oligodendrocytes. These nuclear receptors are recognized as key orchestrators of cellular responses to inflammation and oxidative stress, lipid metabolism, and developmental processes essential for myelin integrity. While accumulating evidence highlights the capacity of PPAR agonists to modulate glial states and support remyelination, knowledge gaps persist regarding their specific mechanisms of action, the interplay between glial subtypes, and the optimization of strategies for therapeutic intervention in complex neurological environments. This Research Topic aims to deepen the mechanistic understanding of how PPAR isoforms regulate glial cell responses and to explore their therapeutic potential in demyelinating and neuroinflammatory disorders. By emphasizing both cell-type-specific actions and shared signaling pathways, the Topic seeks to stimulate the development of innovative approaches that leverage PPAR-targeting compounds to sustain glial homeostasis, mitigate inflammation, and enhance remyelination. The primary objective is to forge clearer links between basic research findings and translational opportunities for preserving white matter integrity. The scope of this Research Topic is limited to studies elucidating the molecular and cellular mechanisms of PPAR signaling in astrocytes, microglia, and oligodendrocytes in relation to demyelinating and neuroinflammatory contexts. To promote focused discussion, we invite articles addressing, but not limited to, the following themes:• Cell-specific effects of PPAR activation in glial populations• Modulation of neuroinflammation and remyelination by PPAR-targeting strategies• Therapeutic opportunities using natural and synthetic PPAR agonists Topic editors: Antonietta Bernardo, Chiara De Nuccio, Roberta Facchinetti Source: https://www.frontiersin.org/research-topics/79937/ppar-signaling-in-glial-cells-mechanisms-and-therapeutic-insights
