Precision immunology and translational advances in rheumatic diseases
Rheumatic diseases represent a diverse group of autoimmune and inflammatory disorders characterized by complex immune dysregulation that affects multiple tissues, including joints, bones, and systemic organs. Despite major therapeutic progress, heterogeneity in disease mechanisms and clinical manifestations continues to hinder patient stratification and optimal treatment outcomes. Recent advances in immunology, single-cell technologies, and multi-omics approaches have begun to unravel the intricate immune networks and molecular pathways underpinning disease heterogeneity. Studies now highlight distinct immune signatures driving different clinical phenotypes and influencing therapeutic responses, yet challenges remain in translating these mechanistic insights into precise, individualized interventions. Although current biologics and small-molecule drugs target major inflammatory pathways, the variability in patient responses underscores the need for refined biomarker-based strategies and better mechanistic understanding to guide therapy. Continued integration of multi-layered immunological data with clinical features is therefore essential to drive forward personalized approaches in rheumatic disease management. This Research Topic aims to advance precision immunology in rheumatic diseases by unifying mechanistic research with translational applications. Specifically, the goal is to delineate immune pathways and regulatory circuits that drive disease onset and progression, identify biomarkers predictive of therapeutic outcomes, and explore novel strategies for patient categorization and targeted treatment. By linking fundamental immunological discoveries to clinical insights, we seek to define how immune heterogeneity informs precision medicine frameworks and potential curative approaches. Key questions include how immune cell interactions and cytokine signaling shape tissue pathology, which molecular signatures serve as actionable biomarkers, and how emerging technologies can refine therapeutic interventions. To gather further insights into the intersection of immune mechanisms and clinical translation, we welcome contributions within the boundaries of rheumatic disease immunopathogenesis and treatment. Submissions may include basic, translational, and clinical investigations using molecular, cellular, and systems-level approaches. We invite articles addressing, but not limited to, the following themes: • Immune cell function and dysregulation in rheumatic diseases• Single-cell and spatial omics technologies for immune profiling • Cytokine signaling networks and inflammatory pathways• Immune regulation of bone and joint pathology (osteoimmunology)• Biomarker discovery, validation, and patient stratification• Translational studies linking immune signatures to treatment response• Novel therapeutic targets and precision medicine strategiesWe welcome original research, reviews, and perspective articles that advance precision-based understanding and management of rheumatic diseases.
