Precision Medicine in Pediatric and Adolescent Sarcomas

Pediatric sarcomas are a significant category within pediatric malignancies, comprising over 20% of solid malignant cancers in children. These cancers are notable for their diverse histological subtypes, originating from mesenchymal cells, and include both soft tissue and bone sarcomas. A critical challenge lies in the varied clinical behavior due to unique genetic and chromosomal characteristics, which complicates diagnosis and treatment. Recent advancements have seen a rise in precision and quality demands for diagnosing these cancers, incorporating techniques such as immunochemistry, FISH, and next-generation sequencing. Despite progress, current diagnostic methodologies still face limitations in effectively addressing the genetic complexity of these cancers. The primary aim of this Research Topic is to explore and highlight how cutting-edge technologies can enhance the understanding and treatment of pediatric sarcomas. By focusing on recent advances in high-throughput genomic and epigenetic techniques, the topic seeks to unveil the genetic and molecular underpinnings that define these tumors. It also seeks to address how integration of artificial intelligence can innovate data analysis and lead to improved diagnostic accuracy and treatment outcomes, thus advancing the field of precision medicine specific to pediatric sarcomas. To gather further insights in the complexity and management of pediatric sarcomas, we welcome articles addressing, but not limited to, the following themes:- Molecular Profiling in Pediatric and Adolescent Sarcomas: Overview of molecular profiling techniques and their role in identifying genetic alterations that drive sarcoma behavior and treatment response.- New Diagnostic Criteria and Laboratory Techniques: Exploration of advanced diagnostic criteria and innovative laboratory techniques that improve the accuracy of pediatric sarcoma classification.- Epigenetic and Methylation Studies in Sarcomas: Examination of epigenetic changes, including DNA methylation patterns, in pediatric bone and soft tissue sarcomas and their role in tumor classification and treatment.- Optic Genome Mapping in Infant Sarcomas: Investigation of optic genome mapping techniques to study the genetic landscape of sarcomas in infancy, providing insights into early cancer development.- Immunochemistry and Precision Medicine in Pediatric Sarcomas: Analysis of immunochemistry approaches and their integration into precision medicine for targeted therapy in pediatric sarcomas.- Advances in Drug Development and Targeted Treatments: Review of the latest developments in drug discovery, targeted therapies, and precision medicine aimed at improving outcomes for pediatric sarcoma patients.- Next-Generation Sequencing in Pediatric Sarcomas: Application of whole-exome sequencing, whole genome sequencing, and RNA-seq to identify molecular signatures and improve personalized treatment strategies.- Clinical Trial Outcomes and Basket Trials in Pediatric Sarcomas: Evaluation of clinical trial results, including basket trials, and their impact on expanding treatment options for pediatric sarcoma patients.- Artificial Intelligence in Pediatric Sarcoma Diagnosis and Treatment: Discussion of AI applications in radiological imaging, genomic data analysis, and treatment decision-making to enhance precision in pediatric sarcoma care. Please note: Manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases, which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.Dr. Jang declared he is affiliated with City of Hope National Medical Center, a private and non-profit institution.
