Precision Medicine in Solid Tumors: Advances, Challenges and Future Outlook

In the current global oncology landscape, solid tumors comprise over 70% of all cancer cases. They are responsible for the majority of cancer-related morbidity and mortality, with the five most common cancer types belonging to this group. Treatment for solid tumors is multidisciplinary, involving a combination of chemotherapy, radiation, surgery, stem cell transplant and immunotherapy. Despite its use, conventional chemotherapy has limitations such as non-specific target engagement, leading to higher morbidity and side effects. Precision medicine, on the other hand, is a novel approach that utilises an individual patient’s genetic, molecular, and clinical profiles to determine treatment. It offers significant benefits such as accurate diagnosis and targeted treatments, reduced side effects, and improved health outcomes for patients. The goal of this Research Topic is to delineate recent advances in precision medicine for solid tumours, including personalised cancer vaccines or immunotherapies, antibody-drug conjugates (ADCs), protein degraders/PROTACs, mutation-targeted therapies, liquid biopsies, and biomarker diagnostics. The topic also encourages scientific discussions on the advantages of precision medicine over conventional chemotherapy, particularly regarding its potential to improve patient health outcomes. Another goal of this Research Topic is to discuss current challenges and the future outlook for personalised medicine in solid tumours to identify areas for further research and improvement. We welcome scientific contributions for this Research Topic, including Original Research, Review, Mini Review, and Perspective articles. Themes include, but are not limited to:1. Novel and current methodologies to identify molecular targets and biomarkers for precision medicine in solid tumors.2. Application of machine learning and artificial intelligence (AI) for diagnostics and treatment prediction. 3. Advances in precision medicine such as immunotherapy, targeted therapy, epigenetic biomarkers, CRISPR-based gene editing, and molecular profiling. Please note: Manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.4. Utilisation and/or integration of next-generation sequencing (NGS), microbiome analysis, omics-based approaches, AI, to identify novel biomarkers and support targeted precision therapies.5. Challenges to the broader application of precision medicine such as socioeconomic and access disparities, technical difficulties, and a lack of public awareness.6. Future opportunities/outlook for precision medicine in solid tumors. Please note: Manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.
