Precision Neuropharmacology in Neurodegenerative Diseases: From Mechanisms to Therapeutics
Neurodegenerative disorders such as Alzheimer’s disease, Parkinson’s disease, amyotrophic lateral sclerosis (ALS), and frontotemporal dementia (FTD) involve distinct but sometimes overlapping disease processes. These include the accumulation of misfolded proteins, such as amyloid-β, tau, α-synuclein, and TDP-43, alongside neuroinflammation, synaptic dysfunction, impaired protein clearance, and mitochondrial or lysosomal deficits. This Research Topic will examine how these mechanisms can guide the discovery and evaluation of treatments, and how molecular, genetic, or biomarker profiles may help identify which interventions are most likely to benefit particular patient groups. We invite preclinical and clinical studies, reviews, and methods papers that link a defined neurodegenerative mechanism to a pharmacological target, the translation of these discoveries into diagnostic and therapeutic strategies, or measurable treatment response. We are particularly interested in work on:- Pharmacological modulation of amyloid-β, tau, α-synuclein, or TDP-43 production, aggregation, spread or clearance- Therapies targeting neuroinflammatory pathways and disease-associated microglial or astrocytic activity- Interventions affecting synaptic function, excitotoxicity, mitochondrial function, autophagy, or lysosomal and proteasomal clearance- Small molecules, biologics, immunotherapies, RNA-based treatments, and gene-targeted approaches, including studies of their mechanisms and pharmacological effects- Bioinformatics and AI/ML methodologies for multi-omics integration, biomarker discovery, and predictive modeling in neurodegeneration- Biomarker- or genotype-informed approaches to patient stratification, treatment selection, and monitoring of target engagement or response- Pharmacokinetics, pharmacodynamics, blood–brain barrier penetration, and drug-delivery strategies that affect central nervous system exposure- Combination or stage-specific treatment strategies, including studies that address interactions between disease-modifying and symptomatic therapies- Translational studies that compare experimental models with human disease or assess why promising candidate therapies do or do not translate into clinical benefit Topic editors: Marios Krokidis, Vittorio Maglione Source: https://www.frontiersin.org/research-topics/86786/precision-neuropharmacology-in-neurodegenerative-diseases-from-mechanisms-to-therapeutics
