Predictive Biomaterial-Integrated In Vitro Platforms for Translational Therapeutic Testing and Delivery Assessment
Biomaterials science is driving innovation in tissue engineering and regenerative medicine by enabling the creation of functional microenvironments that closely replicate the architecture and cues of native extracellular matrices. Hydrogels and three-dimensional scaffolds offer tunable physicochemical and mechanical features, facilitating more physiologically relevant in vitro models for cell behavior and tissue maturation. With the strategic incorporation of micro- and nanoparticles, biomaterials can deliver localized and controlled bioactive signals, while advances in biofabrication and 3D bioprinting provide unprecedented control over the spatial assembly of cells and materials. As a result, biomaterial-integrated microphysiological systems have emerged as sophisticated platforms to emulate tissue-specific microenvironments, facilitating the evaluation of cell-material interactions, maturation processes, and therapeutic responses in a preclinical context. Despite these advancements, challenges remain in establishing in vitro models that not only mimic physiological complexity but also deliver robust predictive and translational value for therapeutic development. This Research Topic aims to advance the development and application of biomaterial-integrated in vitro platforms with high predictive power for translational therapeutic testing and delivery. The central goal is to highlight innovative solutions where engineered hydrogels, scaffolds, and micro- or nanoparticle strategies are employed within advanced microphysiological systems to achieve rigorous assessment of therapeutic efficacy, delivery kinetics, and functional tissue outcomes. The topic invites research that addresses the integration of biomaterial technologies with platforms designed for clinically meaningful readouts, standardized validation, and improved relevance to in vivo and clinical benchmarks. Contributions should demonstrate how new fabrication methods, material properties, or modular system designs can enhance the reproducibility, sensitivity, and translational potential of in vitro regenerative models. The scope of this Research Topic is centered on advances at the intersection of biomaterials engineering, in vitro platform design, and translational therapeutic testing. Submissions should focus on the use of engineered materials to enable predictive, quantitative evaluation of therapeutic delivery and efficacy in tissue-like settings.
