Real-World Data and Real-World Evidence in Lung Cancer, Volume III

This is the third volume of this series collection. Please see the first volume here. Real-World Data and Real-World Evidence in Lung CancerPlease see the second volume here. Real-World Data and Real-World Evidence in Lung Cancer Volume IILung cancer remains one of the most common malignancies worldwide and the leading cause of cancer-related death. Despite substantial advances in diagnosis and treatment, prognosis remains poor, particularly for patients with advanced-stage disease, with 5-year survival varying considerably according to disease stage and geographic region. A substantial proportion of patients are diagnosed with advanced disease and therefore have limited opportunities for potentially curative treatment. Moreover, many patients encountered in routine clinical practice are underrepresented in, or ineligible for, randomized clinical trials. High-quality real-world evidence in advanced-stage lung cancer is therefore essential to complement clinical trial evidence and improve patient outcomes.Although randomized controlled trials (RCTs) remain fundamental for evaluating the efficacy and safety of new interventions, their highly selected populations and controlled settings may leave important gaps in our understanding of treatment effectiveness in routine clinical practice. Real-world data (RWD) and real-world evidence (RWE) can help address these gaps by evaluating treatment effectiveness and safety in broader patient populations, detecting uncommon or long-term adverse events, informing clinical and regulatory decision-making, and identifying patient subgroups that may be more or less likely to benefit from particular therapies. RWD and RWE can also improve our understanding of treatment patterns and outcomes across different geographic, healthcare, and socioeconomic settings. Multicenter real-world studies are particularly encouraged, including well-designed studies of integrative and supportive care approaches used alongside standard systemic therapy, provided that they incorporate rigorous clinical validation and appropriate methods to address confounding and other sources of bias. This Research Topic will focus on high-quality real-world evidence addressing disease and patient characteristics, treatment patterns, therapeutic effectiveness, safety, and long-term outcomes in lung cancer. We welcome Original Research and Review articles. In particular, we encourage prospective clinical studies, multicenter datasets, robust translational investigations, and systematic reviews. Small, single-center retrospective studies will generally fall outside the primary scope of this collection. Topics of interest include, but are not limited to:• Real-world studies of the effectiveness and safety of immune checkpoint inhibitors (ICIs).• Real-world studies of the effectiveness and safety of tyrosine kinase inhibitors (TKIs).• Real-world studies evaluating treatment effectiveness and long-term outcomes in patients with lung cancer, including those with advanced or metastatic (stage IIIB–IV) disease.• Real-world studies evaluating treatment-related toxicity, safety, and long-term adverse events.• Real-world evidence concerning molecular diagnostics, biomarkers, and liquid biopsy in lung cancer.• Real-world studies of radiotherapy outcomes in patients with oligometastatic or oligoprogressive lung cancer.• Comparative and methodological studies examining randomized clinical trials and propensity score–based real-world analyses, including propensity score adjustment, weighting, and matching (PSA/PSM), and their clinical implications.• Multicenter real-world studies of integrative and supportive care interventions delivered alongside standard systemic anticancer therapy, provided that they incorporate rigorous clinical validation and appropriate methods to address confounding, selection bias, immortal time bias, and other potential sources of bias. Please note that manuscripts based solely on bioinformatics, computational analyses, or predictions derived from public databases, without appropriate validation in an independent cohort or biological validation in vitro or in vivo, will not be considered for publication in this Research Topic. The Topic Editors did not declare any potential competing interests in relation to the Research Topic theme.
