Reviews in Systems and Synthetic Immunology
The domain of Systems and Synthetic Immunology is rapidly emerging at the intersection of immunology, systems biology, and engineering, promising profound impacts on both our fundamental understanding of immune processes and the development of innovative medical interventions. Through integrative approaches that combine genomics, computational modeling, cellular engineering, and high-dimensional biological analyses, this field seeks to elucidate the complex networks governing immune responses under both healthy and diseased states. Despite impressive technological advances ranging from genome editing to synthetic cell design, a unified, systems-level perspective on immune regulation remains incomplete. There is significant ongoing debate around optimizing methods for immune modulation, rational vaccine development, and the engineering of next-generation immunotherapies, while gaps persist in our capacity to predict or control emergent immune phenomena.Recent studies in systems and synthetic immunology have achieved numerous breakthroughs, such as the engineering of immune cells for targeted therapies, development of synthetic vaccines, and application of high-dimensional "omics" platforms to map immune system dynamics. Yet, the field continues to face significant questions regarding the translation of these strategies to clinical practice, mechanistic understanding of immune network dysfunction, and the ethical and safety implications of engineered immune interventions. This Research Topic aims to provide a platform for comprehensive and insightful review articles that evaluate emerging concepts, methodologies, and applications within systems and synthetic immunology. The goal is to consolidate advances, identify persisting challenges, and offer fresh perspectives poised to accelerate further discovery. Submissions should address central questions such as: What are the dominant mechanisms underpinning successful immune engineering? How can systems-level data be most effectively leveraged for clinical translation? Where are the greatest unmet needs and opportunities for innovation? We invite contributions that explore, but are not limited to, the following themes:· Engineering biological entities or cells for immune modulation· High-dimensional analyses and modeling of immune system development, dynamics, and function· Systems-level strategies for understanding pathogen, autoimmune, and inflammatory responses· Application of systems biology to therapeutic development for immune disorders· Innovations in vaccine development driven by systems and synthetic approaches· Integration of bioinformatics, nanotechnology, or biomaterials in immunological engineering· Ethical, translational, and safety considerations in the clinical deployment of engineered immune strategiesThis Research Topic welcomes Review, Systematic Review, and Mini Review articles only.
