Revolutionizing Neurological Health: Synaptic Repair and Circuit Remodeling Innovations

Recent advances in neuroscience are reshaping our understanding of how synaptic repair and circuit remodeling influence neurological health. Despite significant progress, many neurological and neuropsychiatric disorders remain difficult to treat, largely due to persistent synaptic dysfunction, maladaptive plasticity, and disrupted neural connectivity. Researchers increasingly recognize that restoring circuit integrity—rather than merely suppressing symptoms—may provide more durable therapeutic benefit. A new generation of strategies is emerging, leveraging engineered synaptic proteins, precision gene editing, activity-dependent circuit modulation, cell-based therapies, and innovative biophysical and biochemical tools to restore or enhance synaptic function and network stability. Advances in imaging and circuit interrogation technologies now enable targeted manipulation of synaptic strength and excitation–inhibition balance across scales. Although early preclinical and clinical studies show promise, major challenges remain in achieving specificity, safety, durability, and effective integration within complex neural circuits. A deeper mechanistic understanding of synaptic and circuit reorganization will be essential for translating these innovations into reliable clinical applications. This Research Topic aims to spotlight pioneering research dedicated to the repair of synapses and the remodeling of neural circuits in the context of neurological disease. Our objective is to foster interdisciplinary discussion on the integration of molecular, cellular, and engineering approaches that enable restoration or regeneration of neural networks. We specifically seek to address outstanding questions such as: Which molecular pathways are most amenable to therapeutic intervention? What are the current limitations of engineered and cell therapy synaptic repair? What are the strategies to modify pathological neuronal circuitry? How can innovative delivery systems and biointerfaces accelerate functional recovery? By focusing on both mechanisms and translation, we hope to advance the development of interventions that provide not only symptomatic relief but also durable improvements in neurological function. This Research Topic welcomes articles that contribute to the advancement of synaptic repair and circuit remodeling. Contributions may range from fundamental mechanistic studies to preclinical and emerging clinical applications but should clearly focus on neurological health. To provide further insights into the field, we invite submissions addressing (but not limited to) the following themes: • Molecular and cellular mechanisms governing synaptic repair • Engineering of synaptic proteins and artificial synapses • Cell therapy alternatives for neuroregeneration • Neuromodulation and neurotechnology for circuit remodeling • Translational strategies bridging laboratory discoveries and clinical interventions We encourage a wide range of submissions, including original research, methods papers, brief research reports, data reports, systematic reviews, mini reviews and perspectives.
