Safety and Efficacy of Gene-Edited Engineering Cell Lines

Gene-edited cell lines represent a transformative frontier in biomanufacturing, particularly for viral vaccine production and therapeutic protein synthesis. Recent innovations in CRISPR/Cas-based systems and other precise genome-editing technologies have enabled targeted manipulation of key cellular traits, including suspension growth, immortalization, and productivity. However, significant challenges persist in ensuring biosafety and genetic stability, especially when transitioning from laboratory-scale systems to industrial bioproduction. Current studies reveal that while genetic modifications can improve adaptability and yield, they may introduce tumorigenic risks or unpredictable genetic drift, limiting large-scale adoption. Although recent research has introduced improved screening and validation methods, a comprehensive framework integrating functional performance with safety evaluation remains insufficiently developed. The field requires a concerted effort to synthesize existing knowledge and establish standardized design and assessment criteria for safe, high-efficiency engineered cell lines. This Research Topic aims to close the gap between advanced gene-editing technologies and their safe industrial implementation. The central objective is to explore strategies that balance high productivity and stability with rigorous biosafety standards. Key questions include how to achieve stable suspension adaptation without compromising genetic integrity, how to immortalize primary cells while suppressing tumorigenic potential, and how to accelerate viral vaccine yield through rational genome modifications. By uniting advances in molecular engineering, cellular biology, and bioinformatics, this initiative seeks to foster the creation of tumor-free, high-performance gene-edited cell lines suitable for scalable biomanufacturing and clinical applications. This Research Topic focuses on the safe development and practical utilization of gene-edited engineering cell lines for biomanufacturing. The scope encompasses both foundational research and applied studies spanning process optimization and biosafety evaluation. To gather further insights into this area, we welcome articles addressing, but not limited to, the following themes:o Gene-editing strategies to promote stable cell suspension and culture scalability.o Genetic and epigenetic mechanisms underlying cell immortalization without tumorigenicity.o Optimization of viral vaccine yield and quality through targeted genomic modifications.o Evaluation and mitigation of biosafety risks in engineered cell lines.o High-throughput screening and validation pipelines for tumor-free engineered cells.o Integration of computational design and multi-omics tools for functional phenotype prediction.o Process engineering and bioreactor systems for large-scale suspension culture.
