Serological signatures in autoimmune diseases: from biomarker stratification to personalized therapeutics
Systemic autoimmune diseases, including systemic sclerosis (SSc), systemic lupus erythematosus (SLE), and rheumatoid arthritis, are defined by profound clinical and pathological heterogeneity that poses persistent challenges to prognostication and therapeutic decision-making. Patient outcomes and treatment responses vary widely even within the same diagnostic entity, creating an urgent unmet need for robust, clinically actionable biomarkers to stratify risk and enable individualized management.Serological signatures, such as autoantibody repertoires, cytokine/chemokine profiles, and circulating immune mediators, represent one of the most accessible and translationally promising tools for decoding the heterogeneity of autoimmune disorders. High-throughput serological profiling and multi-omics integration have recently enabled identification of distinct serological subgroups linked to specific disease trajectories, organ involvement patterns, and therapeutic susceptibility. In SSc, for example, anti-centromere (ACA) and anti-topoisomerase I (anti-Scl-70) antibodies demarcate clinically distinct subsets with divergent prognostic risks, yet the full immunological mechanisms underlying these serological phenotypes remain incompletely characterized. This Research Topic aims to showcase cutting-edge advances in serological signature research across the full spectrum of autoimmune diseases, with a core focus on bridging biomarker discovery and clinical translation. We welcome original research, systematic reviews, and translational studies investigating:- Novel autoantibody specificities- Associations between serum cytokine/metabolomic profiles and disease progression- Multi-omics dissection of immunological pathways linked to distinct serological statuses- The utility of serological markers in predicting therapeutic response. Cohort validation studies and work exploring serological signatures as guides for personalized therapeutic strategies are particularly encouraged. This topic aligns with the mission of the Autoimmune and Autoinflammatory Disorders section, advancing our understanding of immune dysregulation in autoimmunity and accelerating the development of precision medicine approaches.
