Signaling of Innate Immune Sensing: cGAS–STING, Nucleic-Acid Sensors and Inflammasomes in Stress, Metabolism and Disease

Cells constantly sense danger and stress through innate immune signaling. Pattern-recognition receptors and cytosolic sensors, cGAS–STING, RIG-I/MAVS, Toll-like receptors and the inflammasomes, convert cues such as mislocalised DNA/RNA, mitochondrial damage and metabolite shifts into transcriptional and inflammatory programs. These pathways do not act in isolation: nutrient- and energy-sensing signaling (AMPK, mTOR, HIF) and metabolites (itaconate, succinate, lactate, NAD⁺) tune sensor activity, while sensing outputs reshape cellular metabolism. Dysregulated sensing drives sterile inflammation, senescence, autoimmunity, neurodegeneration, infection and cancer, making these nodes prime therapeutic targets. This Research Topic assembles mechanistic and translational work on how innate immune sensing pathways transduce stress and metabolic signals in health and disease, and how they can be pharmacologically tuned. We welcome Original Research, Reviews, Mini Reviews, Methods, Hypothesis & Theory and Perspectives on the following themes but not limited to:o     cGAS–STING signaling: cGAMP second-messenger dynamics, regulation, and crosstalk with interferon and autophagy pathwayso     Nucleic-acid sensing beyond STING: RIG-I/MAVS, TLRs, AIM2 and emerging cytosolic sensorso     Inflammasome assembly, activation and resolution; NLRP3 and metabolic control of inflammasome signalingo     Metabolite and organelle (mitochondria, ER, lysosome) regulation of innate sensingo     Nutrient/energy-sensing cross-talk (AMPK, mTOR, HIF, sirtuins) with immune signalingo     Spatio-temporal and single-cell/spatial approaches, biosensors and computational modeling of sensing dynamicso     Translational and therapeutic modulation (agonists/antagonists, senescence, autoimmunity, infection, cancer immunotherapy)Only manuscripts investigating immunological aspects of inflammation should be submitted to Frontiers in Immunology Topic editors: Zhi-Gang Zhang, Fangfei Li Source: https://www.frontiersin.org/research-topics/84261/signaling-of-innate-immune-sensing-cgas-sting-nucleic-acid-sensors-and-inflammasomes-in-stress-metabolism-and-disease
