Stem Cell-Derived Exosome Cargo: MSC and ADSC Mechanisms in Cartilage, Gut, and Neural Repair
This Research Topic examines the therapeutic mechanisms of MSC and ADSC-derived exosomes across inflammatory and degenerative disease contexts, with a focus on how vesicle cargo, proteins, lipids, and regulatory RNAs, drives tissue-specific repair at the cellular and molecular level. In joint disease, exosomal cargo attenuates chondrocyte apoptosis, promotes extracellular matrix synthesis, and modulates the local inflammatory environment, positioning these vesicles as a defined acellular alternative to direct cell therapy. In inflammatory bowel disease and perianal fistulas, ADSC-derived exosomes activate fibroblast and epithelial repair pathways, restore mucosal integrity, and regulate immune cell responses through cargo-dependent mechanisms that are only beginning to be characterised. In stroke and acute neural injury, MSC exosomes cross compromised blood-brain barrier interfaces, deliver neuroprotective miRNAs, and interact with gut microbiota to support central nervous system recovery, connecting peripheral and central repair through a shared vesicular platform. This collection welcomes original research articles, reviews, mini-reviews, and methods papers across areas including exosome biogenesis and cargo characterisation, tissue-specific repair mechanisms, engineered vesicles for targeted delivery, and the translational challenges of exosome-based therapeutics.
