Systemic Delivery Systems for Leukemia Treatment
Leukemia remains one of the most challenging hematological malignancies, with existing treatments often limited by poor drug bioavailability, systemic toxicity, and the inability to selectively target malignant cells. Systemic delivery systems have emerged as promising tools to overcome these limitations, enabling the targeted and sustained release of therapeutic agents directly to leukemic cells while minimizing off-target effects. These platforms encompass a diverse range of nanocarriers, lipid-based nanoparticles, polymeric systems, and naturally derived extracellular vesicles, each offering unique advantages for drug stability, circulation time, and tissue-specific targeting.Among these, membrane-bound vesicles, such as exosomes and microvesicles, are gaining attention as biomimetic delivery vectors. Their endogenous origin allows them to evade immune detection, cross biological barriers, and deliver both small molecules and macromolecular drugs with high specificity. By engineering vesicle surfaces with leukemia-targeting ligands or antibodies, researchers can enhance therapeutic precision and reduce systemic toxicity. Combining vesicle-based systems with nanotechnology further expands their potential, enabling controlled drug release and real-time tracking through imaging agents.Advances in systemic delivery strategies could also facilitate the use of novel therapeutic modalities for leukemia, including RNA-based drugs, CRISPR gene-editing components, and immune-modulating agents. Ultimately, integrating bioengineering, materials science, and oncology may lead to highly personalized leukemia treatments with superior efficacy and safety profiles. This research topic will focus on:• Development of nanocarrier platforms for systemic delivery in leukemia.• Engineering membrane vesicles for targeted anti-leukemic therapy.• Improving pharmacokinetics and bioavailability of leukemia drugs.• Strategies to reduce systemic toxicity and off-target effects.• Integration of imaging agents for real-time tracking of therapeutic delivery.• Application of vesicle-nanoparticle hybrids for enhanced treatment efficacy.• Exploring RNA and gene-editing therapeutics delivered via systemic platforms Topic editors: Ashanti C.
