Systemic Lipid Metabolism and Multi-Organ Crosstalk: Orchestrating Cardiometabolic Health and Endocrine Signaling
Lipid metabolism in cardiometabolic health is increasingly recognized as an interdependent, multi-organ process, rather than a function confined to individual tissues. Groundbreaking research over the past decade has revealed that organs such as adipose tissue, liver, muscle, and the heart are in constant metabolic conversation, facilitated by a network of lipid-derived signaling molecules and other endocrine mediators. This paradigm shift highlights the complexity of systemic energy regulation and vascular health, moving beyond reductionist views that once compartmentalized metabolic pathways. New studies demonstrate that disruptions to this coordinated communication—due to lifestyle factors, genetic predispositions, or environmental influences—can propagate a cascade of metabolic dysfunctions, culminating in conditions such as insulin resistance, non-alcoholic fatty liver disease (NAFLD), atherosclerosis, and heart failure. Despite these advances, significant gaps persist in mapping the precise molecular mechanisms that drive organ-specific contributions and their dynamic interactions, particularly under pathophysiological conditions. This Research Topic aims to elucidate the mechanisms through which lipid metabolism serves as a unifying “language” orchestrating crosstalk among key organs—including the heart, liver, adipose tissue, muscle, and gut—within the context of cardiometabolic disorders. Specifically, the objective is to redefine the heart’s role from a passive recipient of metabolic dysfunction to an active participant in sensing, integrating, and responding to systemic lipid fluxes. Central questions involve identifying the molecular circuits of inter-organ communication, clarifying how lipid-derived signals coordinate homeostasis or pathology, and exploring therapeutic interventions that target these axes to ameliorate or reverse disease progression. The scope of this Research Topic encompasses mechanistic, translational, and clinical studies investigating how systemic lipid handling impacts inter-organ signaling and cardiometabolic outcomes. The focus is on integrative models and multi-omics approaches that unravel the complexity of endocrine lipid networks, while clinical studies are encouraged to bridge experimental insights with patient phenotypes and outcomes.
