Systems Biology of the Microbiome for Targeted Therapeutic Design and Personalized Medicine

The field of integrative systems microbiome biology is reshaping our understanding of human health and disease by highlighting the pivotal role of microbial communities in regulating host physiology. Recent advances show that the microbiome—comprising bacteria, fungi, viruses, and archaea—is acquired at birth and dynamically evolves through environmental exposures, diet, and antibiotic use. These diverse communities modulate host metabolism, immune function, gene expression, and broader phenotypic traits. Current research reveals that disturbances in the microbiome, known as dysbiosis, underlie a range of acute and chronic conditions, from gut diseases and metabolic syndromes to neurological disorders and cancer. Despite the proliferation of studies associating the microbiome with disease, most investigations remain correlative, lacking in mechanistic clarity, reproducibility, and translational clinical application. Multi-omics technologies—metagenomics, metaproteomics, metabolomics, and metatranscriptomics—are increasingly employed to bridge these gaps and provide comprehensive systems-level insights into microbial functions and host interactions. This Research Topic aims to accelerate the transition from descriptive microbiome studies to truly translational outputs in precision medicine. By bringing together advances in multi-omics, computational modeling, and targeted therapeutic strategies, the topic seeks to address critical questions such as: What are the molecular mechanisms driving host-microbiome interactions in disease? How can multi-omics data be integrated to identify actionable therapeutic targets? Which approaches best translate microbiome insights into diagnostics, patient stratification, and personalized interventions? This effort aspires to generate robust, reproducible science that advances mechanistic understanding and supports the clinical translation of microbiome research. To gather further insights into the boundaries of integrative microbiome systems biology for translational and precision medicine, we encourage submissions that focus on multi-omics analyses, innovative data integration, and the development of therapeutic strategies. We welcome articles addressing, but not limited to, the following themes:Integrative multi-omics studies (transcriptomics, proteomics, genomics, metabolomics) of host-microbiome interactions in disease mechanismsTranslation of microbiome research into therapeutic targets and strategies, with an emphasis on metabolic pathway modulationApplication of computational and systems biology, including machine learning, artificial intelligence, and network analysis to uncover mechanistic insights and therapeutic opportunitiesCRISPR-based and other precision editing methods for modulating the structure and function of microbial communitiesApproaches to overcoming challenges of interindividual variability, reproducibility, and standardization in clinical microbiome research Topic editors: Palanichamy Esakkiraj, SAMPATHKUMAR RANGANATHAN, Umashankar Vetrivel, Karthik Maruthan, Sangavi P Source: https://www.frontiersin.org/research-topics/76242/systems-biology-of-the-microbiome-for-targeted-therapeutic-design-and-personalized-medicine
