Targeted Drug Delivery at the Interface of Angiogenesis and Immune Modulation in Cancer
The crosstalk between angiogenesis and immune regulation is a defining feature of the tumor microenvironment (TME), influencing both disease progression and therapeutic responsiveness. Abnormal and leaky tumor vasculature not only supports tumor growth and metastasis but also creates hypoxic, immunosuppressive niches that hinder immune cell infiltration and function. While anti-angiogenic and immunotherapeutic agents have individually shown promise, their clinical efficacy remains limited by poor tumor penetration, transient vascular normalization, and systemic toxicity.Emerging drug delivery technologies, including nanoparticles, liposomes, polymeric systems, exosomes, and stimuli-responsive carriers, offer unprecedented opportunities to spatially and temporally modulate the angiogenesis–immunity axis. By improving drug accumulation in the tumor, enabling co-delivery of angiogenesis modulators and immunotherapeutics, or leveraging vascular normalization windows, these approaches aim to overcome the physical and immunological barriers of the TME.This Research Topic seeks to advance the mechanistic and translational understanding of drug delivery strategies that exploit angiogenic and immune pathways to enhance cancer treatment outcomes. Submissions may include preclinical or clinical studies, computational modeling, or technology-focused innovations that address the design, delivery, and therapeutic optimization of agents acting on this dual axis.We welcome articles addressing, but not limited to, the following areas:- Drug delivery strategies targeting angiogenic pathways (e.g., VEGF/VEGFR, ANG–TIE) to reshape the tumor vasculature and improve immune infiltration- Nanocarriers and biomaterials for the co-delivery of anti-angiogenic and immunomodulatory agents- Temporal and spatial control of drug release to align with vascular normalization and immune activation windows- Imaging and biomarker-based tracking of vascular and immune responses to delivered therapies- Overcoming tumor heterogeneity and resistance mechanisms through adaptive or smart delivery systems- Computational and systems approaches to optimize dosing, scheduling, and patient stratification in angio-immuno drug delivery- Clinical translation and safety evaluation of combination delivery platforms targeting the angiogenesis–immune interfacePlease note that manuscripts consisting solely of bioinformatics or computational analysis of public omics databases that are not supplemented by relevant functional validation (clinical cohort or biological validation in vitro or in vivo) are out of scope for this Research Topic.
