Targeting inflammasome pathways for precision treatment in allergy and asthma
The field of allergy and asthma research has increasingly recognised the inflammasome as a central player in orchestrating immune responses and tissue inflammation. Inflammasomes, particularly NLRP1 and NLRP3, act as molecular sensors that detect cellular stress, pathogens, and environmental allergens, initiating cascades that drive cytokine release and immune cell activation. Recent studies have emphasised their involvement in asthma and atopic dermatitis, where environmental triggers such as rhinoviruses and Staphylococcus aureus colonisation can amplify inflammatory signalling. While conventional allergy and asthma research has primarily focused on T helper cell imbalances and airway remodelling, emerging evidence suggests that dysregulated inflammasome activation may be a missing link in understanding chronic inflammation and the severity of exacerbations. Despite these advances, the pathways linking inflammasome activation to specific allergic phenotypes, genetic susceptibilities, and environmental exposures remain incompletely understood, necessitating a multidisciplinary approach to unravel these mechanisms. This Research Topic aims to elucidate the molecular and immunological mechanisms underlying inflammasome activation in allergic and asthmatic diseases, with a focus on defining its role in pathogenesis and in therapeutic modulation. By synthesising evidence from basic science, translational research, and clinical studies, this initiative seeks to determine how inflammasomes influence immune regulation, epithelial barrier function, and cytokine signalling. Core objectives include characterising the interplay between genetic variants of inflammasome components and disease susceptibility, exploring the dynamics of inflammasome triggering in allergic inflammation, and evaluating the feasibility of inflammasome-targeted interventions as next-generation therapies for asthma and related allergic disorders. To gain further insights into inflammasome-driven mechanisms in allergy and asthma research, we welcome articles addressing, but not limited to, the following themes:- Molecular and cellular mechanisms of NLRP1 and NLRP3 inflammasome activation in allergic diseases- Interaction of inflammasome pathways with environmental allergens, respiratory viruses, and microbial colonisation- Genetic and epigenetic regulation of inflammasome components and their link to asthma and atopy- Crosstalk between inflammasome signalling and adaptive immune responses in airway and skin inflammation- Preclinical and clinical evaluation of inflammasome-targeted therapeutic strategies- Role of inflammasome-associated cytokines (e.g., IL-1β, IL-18) in the progression and exacerbation of asthma- Integration of molecular profiling and biomarker discovery for precision medicine approachesAll article types are welcome, including original research, systematic reviews, mini-reviews, letters, perspectives, clinical case studies, and FAIR² Data Articles.
