Targeting Metabolic Factors of Antimicrobial Resistance

Antimicrobial resistance among pathogens continues to affect global public health, reducing the effectiveness of existing medicines and increasing the prevalence of infectious diseases. While genetic factors such as mutations and horizontal gene transfer are well-established contributors to resistance, there is emerging recognition that bacterial metabolism plays a major role in developing antimicrobial responses. Under stress conditions, bacteria can dynamically reconfigure their metabolic pathways, carbon metabolism, energy production, cellular redox balance and maintain cellular homeostasis. Altered metabolic states can influence oxidative stress responses, signaling and persistence, enabling bacterial populations to resist antibiotic drugs without necessarily developing long-term genetic resistance. Thus, understanding these metabolic adaptations is essential for developing more effective strategies to combat antibiotic tolerance, which prolongs the time for genetic diversification resulting in the emergence of resistance in bacterial populations. This Research Topic will highlight recent developments in our knowledge of how metabolic processes underlie antibiotic resistance and tolerance and investigate novel approaches that target these pathways.
