Targeting Neuroinflammation: Mechanisms, Pathways and Therapeutic Opportunities

Neuroinflammation is a critical pathological process at the intersection of immunology and neuroscience. It is now established as a key driver in the onset and progression of a wide range of central nervous system (CNS) disorders, including Alzheimer’s and Parkinson’s disease, multiple sclerosis, traumatic brain injury, and depression. The complex interplay between activated glial cells, inflammatory signaling, and neuronal function presents both a significant challenge and a compelling opportunity for therapeutic intervention. Despite significant advances in understanding these mechanisms, effective pharmacological treatments targeting neuroinflammation remain scarce outside of broad immunosuppressive approaches. With the global burden of neurological disorders rising, the need for targeted, CNS-active anti-inflammatory therapies has never been more pressing. While our understanding of the fundamental mechanisms of inflammation has advanced, translating these insights into effective neuropharmacological agents remains a major hurdle. The unique environment of the CNS, protected by the blood-brain barrier, demands innovative strategies for both drug design and CNS target engagement. This Research Topic aims to connect these fields by showcasing cutting-edge research that translates mechanistic insights into novel drug development for inflammation-related diseases of the nervous system. We welcome submissions that provide in-depth knowledge of how to pharmacologically target neuroinflammatory processes. The scope is multidisciplinary and includes, but is not limited to:•Molecular and Cellular Mechanisms: Investigation of the molecular mechanisms regulating the activation and resolution of inflammatory responses in microglia and astrocytes, and the subsequent impact on neuronal function, synaptic plasticity, and neurodegeneration.•Novel Drug Targets and Pathways: Identification and validation of new therapeutic targets within inflammatory pathways (e.g., the inflammasome, cytokine signaling) and their role in the context of specific neurological diseases.•Neuropharmacology and Drug Development: The design, synthesis, and evaluation of CNS-penetrant small molecules, biologics, and other therapies, with a focus on their pharmacokinetics, pharmacodynamics, and ability to modulate neuroinflammatory outcomes in relevant models.•Clinical Neuropharmacology: Translational and clinical studies on anti-inflammatory strategies in neurological and psychiatric disorders, including biomarkers, treatment response, and risk factors. This Research Topic supports SDG 3: Good Health and Well-being by advancing therapeutic strategies for CNS disorders.
