The Evolving Tumor-Immune Gene Mutations Interface: Novel Biomarkers for Response, Resistance, and Immunotherapeutic Strategies
The advent of immunotherapy, particularly immune checkpoint inhibitors (ICIs), has revolutionized oncology. However, a significant proportion of patients exhibit primary resistance, while others develop acquired resistance after an initial response. This heterogeneity underscores the limitations of current biomarkers, such as PD-L1 expression and tumor mutational burden (TMB), which lack sufficient predictive precision. The tumor-immune interface is not static but a highly dynamic ecosystem that evolves under therapeutic pressure and disease progression. A deeper understanding of this evolving crosstalk is critical to decipher the mechanisms of response and resistance. Recent technological advances, including single-cell multi-omics, spatial transcriptomics, and high-dimensional immune profiling, now allow us to dissect this complexity with unprecedented resolution. This Research Topic aims to compile cutting-edge research on novel biomarkers derived from the tumor microenvironment and peripheral systems. We seek studies that identify and validate dynamic predictive and prognostic signatures, elucidate mechanisms of immune escape, and translate these findings into strategies to overcome resistance. By integrating basic science with translational and clinical research, this Topic will drive the development of next-generation tools for patient stratification and personalized immunotherapeutic interventions. This Research Topic aims to compile high-impact research that elucidates the dynamic mechanisms at the tumor-immune interface to discover and validate novel biomarkers. We welcome studies leveraging cutting-edge technologies (e.g., single-cell omics, spatial profiling, AI) to define predictive signatures for therapy response, resistance mechanisms, and immune-related adverse events. Our goal is to foster a multidisciplinary dialogue that translates mechanistic insights into clinically actionable strategies, ultimately advancing personalized immunotherapy and improving patient outcomes. This Research Topic welcomes original research, reviews, and methodological advances that explore the dynamic tumor-immune interface to discover and validate novel biomarkers. Specifically, we encourage submissions on:• Mechanistic and Technology-Driven Studies: Research utilizing single-cell/spatial omics, liquid biopsy, advanced imaging, and AI to identify biomarkers predictive of immunotherapy response, resistance (primary/acquired), or immune-related adverse events.• Translational and Clinical Research: Studies validating novel cellular, molecular, or digital biomarkers in preclinical models or clinical cohorts, with a focus on their utility for patient stratification, combination therapy design, or monitoring treatment efficacy.• Therapeutic Strategies: Research proposing biomarker-guided strategies to overcome resistance, such as targeting novel immune checkpoints, metabolic pathways, or the immunosuppressive microenvironment. We aim to foster a multidisciplinary forum bridging immunology, cancer biology, and clinical oncology. All submissions must present robust, hypothesis-driven science and adhere to the highest ethical and reporting standards. Please Note: Manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases, without robust validation such as clinical cohort studies or biological validation, are out of scope for this topic.
