The multifaceted roles of noncoding RNAs in neuroinflammatory responses

Neuroinflammation is a critical process implicated in the pathogenesis and progression of a wide spectrum of central nervous system (CNS) disorders, including neurodegenerative diseases, autoimmune conditions, and acute injury. Recent advancements have highlighted the pivotal regulatory functions of noncoding RNAs (ncRNAs)—encompassing microRNAs, long noncoding RNAs, and circular RNAs—in orchestrating inflammatory responses within the CNS. These ncRNAs modulate key signaling pathways such as NF-κB, JAK/STAT, and NLRP3, influencing transcriptional activity, mRNA stability, and cellular communication between neuronal and glial populations. While growing evidence underscores their involvement in both the amplification and resolution of inflammation, significant questions remain about the context-specific actions, regulatory networks, and interplay among distinct ncRNA species.Emerging studies have begun to unravel how the modulation of ncRNAs can impact disease onset, progression, and recovery in various neurological conditions, with some investigations demonstrating promising therapeutic effects in preclinical models. However, the intricacies of ncRNA-mediated mechanisms are far from fully understood, and the translational potential of ncRNA-targeted interventions remains underexplored. There is a pressing need for comprehensive research to delineate the mechanisms by which ncRNAs interface with immune signaling components and to identify new targets for therapeutic modulation in the context of neuroinflammation. This Research Topic aims to advance understanding of how noncoding RNAs regulate neuroinflammatory processes and their implications for disease progression and treatment. We seek to elucidate the cellular and molecular mechanisms by which ncRNAs govern inflammatory signaling, their roles in neuro–immune interactions, and the potential for ncRNA-based strategies to ameliorate neuroinflammatory conditions. By fostering contributions on both mechanistic insights and translational applications, we hope to catalyze the development of innovative approaches to diagnose, monitor, and treat CNS inflammation. This Research Topic encompasses studies addressing the regulatory actions of ncRNAs in the CNS, with a focus on their influence over neuroinflammation, neural–immune interactions, and disease outcomes.
