Translational Perspectives in Pediatric Liver Diseases: Bridging Pathophysiology, Diagnosis and Therapeutic Interventions

Pediatric liver diseases comprise a diverse and complex spectrum of congenital, metabolic, inflammatory, and neoplastic conditions that can lead to progressive liver injury, fibrosis, and organ dysfunction. Despite advances in experimental research and clinical management, these disorders remain associated with substantial morbidity and long-term complications in pediatric populations. A central challenge in pediatric hepatology lies in the marked heterogeneity of disease presentation and progression, with clinical trajectories often difficult to anticipate. Although significant progress has been made in identifying molecular pathways involved in liver injury, including immune-mediated processes, metabolic dysregulation, and fibrogenic signaling, these mechanisms are often studied in isolation and are not consistently translated into clinically meaningful approaches in pediatric populations. This Research Topic aims to advance the integration of mechanistic and clinical knowledge in pediatric liver diseases through a multidisciplinary and translational framework. We seek contributions that bridge experimental and clinical perspectives, fostering a more comprehensive understanding of disease processes and their implications for diagnosis, prognosis, and treatment. By connecting biological mechanisms with clinical application, this topic aims to support improved strategies for risk stratification, therapeutic decision-making, and patient management. We welcome submissions addressing a broad range of topics in pediatric liver diseases, with particular emphasis on studies that explore the interplay between cellular and molecular mechanisms, experimental models, and clinical manifestations. Contributions integrating multiple levels of investigation, from mechanistic understanding to clinical outcomes are especially encouraged.Submissions may include, but are not limited to:• Mechanisms of liver injury and disease progression, including inflammatory, immune-mediated, metabolic, and fibrogenic pathways, as well as cellular interactions within the hepatic microenvironment• Extrahepatic and systemic manifestations of pediatric liver diseases• Experimental and preclinical models and their translational relevance• Strategies for early diagnosis and disease detection, including biomarkers, imaging, and non-invasive assessment approaches• Clinical heterogeneity, patient stratification, and determinants of disease progression and therapeutic response• Pharmacological, supportive, and interventional therapeutic approaches, including clinical decision-making strategies• Surgical management, including biliary procedures, pediatric liver transplantation, and post-transplant adaptation• Pediatric liver tumors and mechanisms of tumorigenesis Topic editors: Josiane de Oliveira Goncalves, Ana Cristina Aoun Tannuri, Mohamed El Guindi Source: https://www.frontiersin.org/research-topics/80754/translational-perspectives-in-pediatric-liver-diseases-bridging-pathophysiology-diagnosis-and-therapeutic-interventions
