Trauma-induced immune dysfunction: mechanisms, biomarkers, and therapeutic targets
Trauma, especially severe trauma, triggers an immediate and massive release of Damage-Associated Molecular Patterns (DAMPs), rapidly activating multiple innate immune pathways. If left uncontrolled, this instant inflammatory response drives a systemic cascade that ultimately leads to trauma-induced organ failure, including Acute Respiratory Distress Syndrome (ARDS), Acute Kidney Injury (AKI), and Multiple Organ Failure (MOF). Furthermore, early innate immune activation does not occur in isolation; it cross-talks extensively with other critical systems, particularly the coagulation and complement cascades, as well as endothelial cells. This complex interaction generates inflammatory mediators, reactive oxygen species (ROS), and ischemia, exacerbating cellular and tissue damage and progressively worsening clinical outcomes. Therefore, understanding trauma-induced immune dysfunction is fundamental. Deciphering these early mechanisms is critical for developing targeted therapies that can prevent or mitigate trauma-induced organ failure, ultimately saving the lives of both combat casualties and civilian trauma patients. This Special Issue aims to compile a collection of high-impact original research and review articles exploring the diverse mechanisms underlying trauma-induced immune dysfunction. We seek to feature cutting-edge research that bridges the gap between fundamental pathogenesis, molecular discoveries, and the identification of novel therapeutic targets. By focusing on innate immune responses, signaling pathway interactions, cellular crosstalk, and translational applications, we invite submissions including, but not limited to:Systemic Pathophysiology: Innate immune signaling and systemic pathways leading to trauma-induced organ injury and mortality.Pathway Interactions: Synergistic crosstalk between innate immune pathways following traumatic injury.Molecular & Cellular Triggers: The roles of endothelial glycocalyx shedding, complement, inflammatory cascades, and DAMPs in initiating immune dysfunction.Diagnostics & Prognostics: Identification of biomarkers for diagnosis and prognosis, alongside novel technologies for predicting clinical outcomes.Therapeutic Interventions: Pre-clinical studies in animal models to mitigate organ failure and clinical trials evaluating novel therapeutics for trauma-induced injury.
