Tumor associated neurobehavioral dysfunction in glioblastoma: reviewing mechanisms, models, and therapeutic targets

Glioblastoma (GBM) is not only a highly aggressive brain tumor with poor prognosis but also a major contributor to neurobehavioral dysfunction, profoundly impacting cognition, emotion, and behavior, including impairments in memory, attention, language, and executive function. Despite growing recognition of these effects, the complex interactions between tumor biology and neural circuits remain poorly understood, and effective strategies to preserve neurocognitive function are limited. This Research Topic aims to integrate mechanistic insights, experimental models, and translational approaches to review and advance our understanding of tumor-associated neurobehavioral dysfunction in GBM. We particularly welcome full-length, mini, and systematic review articles exploring tumor-associated neurobehavioral dysfunction in glioblastoma, including underlying mechanisms, experimental models, and therapeutic opportunities. Topics of interest include, but are not limited to:1. Mechanistic pathwayso Tumor–neuron synaptic interactions and their impact on neural circuit function.o Neuroinflammatory processes and neurotransmitter alterations underlying cognitive, emotional, and behavioral changes.o The role of the tumor microenvironment, including glial remodeling, metabolic rewiring, immune cell dynamics, and stress-related neuromodulators.2. Experimental and computational modelso In vivo and in vitro models capturing neurobehavioral readouts in GBM.o Advanced approaches such as organoids, microfluidic systems, NAMs (new approach methodologies), and in silico/AI-based simulations.o Opportunities and limitations in modeling tumor-associated neurobehavioral symptoms (TANs).3. Therapeutic targets and interventionso Strategies to preserve neurocognitive function or modulate aberrant neural circuits.o Approaches to disrupt tumor–neuron signaling pathways or alleviate TANs.4. Translational and clinical perspectiveso Biomarkers, neurocognitive assessments, and imaging approaches for monitoring behavioral impairments.o Patient-reported outcomes and integrative strategies for linking preclinical findings to clinical interventions.By bridging neuro-oncology and behavioral neuroscience, this collection seeks to provide a comprehensive synthesis of current knowledge and identify emerging opportunities for intervention to improve patient quality of life.
