Unleashing the Microbiome’s Arsenal: Innovations in Colonization Resistance for Pathogen Control
The field of microbiome research has uncovered the extraordinary defensive capabilities of the resident microbial community in protecting hosts from pathogenic invasion—a phenomenon termed colonization resistance. Mounting evidence highlights that the delicate balance and composition of these microbial populations serve as a biological barrier, limiting the establishment and proliferation of harmful pathogens. Disruption to this microbial harmony, frequently prompted by factors such as antibiotic use, illness, or lifestyle changes, can compromise this innate defense, rendering hosts susceptible to opportunistic and antibiotic-resistant infections. Recent studies reveal that the microbiome leverages a diverse set of mechanisms—including bacteriophages, antimicrobial peptides, inhibitory metabolites, and postbiotics—to actively deter pathogen colonization. However, significant knowledge gaps persist regarding the precise interplay among these microbial components, the conditions that impair their effectiveness, and the strategies by which this protective shield can be therapeutically harnessed or restored. This Research Topic aims to advance our understanding of how the microbiome’s intrinsic “natural arsenal” can be leveraged to restore and enhance colonization resistance. It seeks to gather novel research and comprehensive reviews that dissect the functions, interactions, and synergies among bacteriophages, antimicrobial peptides, inhibitory metabolites, and postbiotics. By illuminating these biological tools and their integration within host defense systems, the Topic intends to identify targeted interventions for combating major pathogens—such as C. difficile, VRE, N. gonorrhoeae, Salmonella, and E. coli—while preserving the delicate balance of beneficial microbial communities. Moreover, it encourages exploration of diet-mediated modulation of microbial defense pathways, aiming to deliver translational insights for innovative prevention and treatment strategies. This Research Topic welcomes investigations spanning mechanistic, translational, and applied research focused on microbial solutions to reinforce host colonization resistance. Relevant submissions may address any body site (e.g., gut, skin, urogenital tract), but must center on endogenous microorganisms or their products. We invite articles addressing, but not limited to, the following themes:The role and therapeutic application of bacteriophages in limiting pathogen colonizationDiscovery, mechanisms, and clinical potential of antimicrobial peptides and bacteriocinsInhibitory metabolites, such as short-chain fatty acids and modified bile acids, in shaping pathogen resistanceMulti-modal and synergistic interactions between microbial agents (e.g., phage and postbiotic combinations)Maintenance and restoration of physical mucosal barriers by microbial productsDiet, prebiotics, and host-derived factors influencing the microbiome’s defensive repertoire Topic editors: Abdallah Abdelsattar, Ahmed Abouelkhair DVM, MS, DACVM, Roberto Mazzoli Source: https://www.frontiersin.org/research-topics/82218/unleashing-the-microbiomes-arsenal-innovations-in-colonization-resistance-for-pathogen-control
