Unlocking the Metabolic Dynamics of Tumor Microenvironment through Radiotherapy: Bridging Pre-Clinical Insights to Clinical Applications

Radiotherapy stands as a cornerstone in cancer treatment, leveraging ionizing radiation to target and eradicate tumor cells. However, beyond its direct cytotoxic effects, emerging evidence underscores its profound impact on the metabolic milieu of the tumor microenvironment (TME). This dynamic interplay orchestrates a complex array of metabolic reprogramming within cancer cells and surrounding stromal components, influencing tumor progression, therapeutic response, and disease recurrence. Understanding the intricate metabolic alterations induced by radiotherapy presents a pivotal avenue for optimizing treatment strategies and overcoming therapeutic resistance. Pre-clinical models have unraveled key molecular mechanisms underlying these metabolic adaptations, yet translation to clinical practice remains a challenge. Addressing this gap requires a comprehensive exploration of radiotherapy-induced metabolic remodeling across diverse tumor types and clinical settings. In this Research Topic, we welcome submissions of original research articles, reviews, clinical trials, translational studies, and perspectives that contribute to our understanding of how radiotherapy-based treatments organize the TME metabolism, with a focus on translating findings from pre-clinical models to clinical practice. This Research Topic invites contributions that investigate the effects of radiotherapy on the TME metabolism across the translational spectrum. Subtopics of interest include, but are not limited to:1. Pre-clinical models, such as 2D models and 3D models for studying radiotherapy-induced metabolic changes in the TME.2. Mechanistic insights into the role of radiotherapy-related metabolic change in modulating immune responses, angiogenesis, and stromal components in TME.3. Clinical imaging techniques for assessing radiotherapy-induced alterations in the TME.4. Biomarkers predictive of TME metabolism response to radiotherapy-based treatments.5. Clinical trials evaluating novel radiotherapy strategies targeting the TME metabolism.Manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases which are not accompanied by robust and relevant validation (clinical cohort or biological validation in vitro or in vivo) are out of scope for this topic.
