Unraveling DNA Molecular Structures and Their Epigenetic Implications: Advances in Spectroscopic and Biophysical Approaches.

This special topic will explore the diverse structural conformations of DNA, such as G-quadruplexes, Z-DNA, triplex DNA, and other non-canonical structures, and their critical roles in gene regulation, chromatin organization, and epigenetic modifications. Since DNA structure is inherently linked to epigenetic mechanisms, including DNA methylation and histone modifications, studying these molecular conformations can provide deeper insights into fundamental biological processes and disease mechanisms.A key aspect of this special topic is the integration of advanced analytical techniques, particularly spectroscopic and imaging approaches with robust data interpretation and statistical rigor, in studying DNA molecular structures. In recent years, methods such as surface-enhanced Raman spectroscopy (SERS), tip-enhanced Raman spectroscopy (TERS), and single-molecule fluorescence spectroscopy have enabled unprecedented sensitivity and specificity in detecting DNA conformational changes and epigenetic modifications at the molecular level. Furthermore, the application of AI-driven spectral analysis and machine learning models is enhancing data interpretation, offering new perspectives in DNA structural studies. This topic will encourage contributions that highlight:• The role of DNA structural variations in epigenetic regulation and disease mechanisms, supported by quantitative data analyses.• Advances in Raman spectroscopy (SERS, TERS), NMR, cryo-EM, and other high-resolution techniques for studying DNA conformations, with an emphasis on studies that apply these approaches to biological questions and statistically evaluate the findings.• Computational and AI-driven methods for DNA structure prediction and spectral analysis, including validation and performance assessment.• The impact of DNA structural polymorphism on gene expression and chromatin accessibility, defined through rigorous experimental design and appropriate statistical analysis.• Emerging applications of spectroscopic techniques in detecting epigenetic biomarkers, illustrated with relevant experimental or computational results.Through this topic, we aim to advance both methodological innovation and biological understanding in the field. We particularly welcome studies that thoughtfully combine advanced techniques and quantitative analysis to reveal new insights into DNA structure and function.
