Unraveling the Immunopathogenic Roles of Gut Microbiota in Lupus: From Mechanistic Insights to Targeted Therapeutic Strategies

The gut microbiome, a complex community of microorganisms residing in the gastrointestinal tract, plays a crucial role in maintaining immune system balance. Increasing evidence suggests a strong association between alterations in gut microbiota composition known as dysbiosis and the development of autoimmune diseases. A healthy microbiome helps regulate immune tolerance and prevents inappropriate immune activation against the body’s own tissues. However, when this balance is disrupted by factors such as diet, infections, antibiotics, or genetics, it can lead to immune dysregulation and chronic inflammation. Dysbiosis has been implicated in several autoimmune disorders, including rheumatoid arthritis, type 1 diabetes, multiple sclerosis, and systemic lupus erythematosus. The interplay between the immune system and specific microbial species antigens, toxins or their metabolites can either promote tolerance or trigger pathogenic immune responses, highlighting the microbiome’s potential as both a biomarker and therapeutic target in Lupus prevention and intervention. This Research Topic welcomes contributions that investigate the role of gut microbiota dysbiosis in modulating immune regulation and driving disease progression in lupus. Submissions employing innovative methodologies such as microbial genomic identification, microbiota transfer in germ-free animal models, metabolomic analysis and pathways-based studies are particularly encouraged to elucidate the complex mechanisms governing microbiota–immune system interactions. These efforts align with the journal’s mission to advance our understanding of immunopathogenesis and foster the development of novel therapeutic strategies in Lupus.
