Unraveling the Intricate Nexus: Pancreatic Cancer in the Context of Metabolic Syndrome, Diabetes - Associated Molecular Signatures, and Endocrine Signaling Cascades

Pancreatic carcinoma is recognized as one of the most aggressive and challenging-to-treat malignancies. Recently, research has revealed substantial evidence connecting pancreatic carcinoma to systemic metabolic disorders, notably metabolic dysregulation syndrome. This syndrome, characterized by abdominal obesity, hyperglycemia, hyperlipidemia, and hypertension, is identified as a significant risk factor for pancreatic carcinogenesis. Chronic inflammation and insulin resistance, often resulting from metabolic syndrome, can activate oncogenic pathways, promoting tumor growth. Additionally, lipid metabolism dysregulation can reshape the tumor microenvironment, aiding cancer cell survival and invasion. Furthermore, diabetes mellitus is intricately linked to pancreatic carcinoma, with diabetes-associated cancers presenting distinct molecular signatures. Dysregulation in endocrine signaling pathways profoundly affects every phase of pancreatic carcinoma, influencing cancer cell behavior and their interaction with surrounding stromal and immune cells. These insights underscore the necessity for exploring these metabolic associations to develop improved therapeutic strategies. This Research Topic aims to delve into the intricate relationships among pancreatic carcinoma, metabolic dysregulation syndrome, diabetes-associated molecular features, and endocrine signaling transduction cascades. By bridging insights from basic, translational, and clinical research, the objective is to unravel the underlying molecular mechanisms of pancreatic carcinoma development. There is a focus on discovering novel diagnostic biomarkers and therapeutic targets for improved early detection and personalized treatment. Additionally, this topic seeks to bridge basic scientific research with clinical applications, translating research findings into tangible benefits for pancreatic carcinoma patients. To gather further insights into pancreatic carcinoma in the context of metabolic syndromes and diabetes, we welcome articles addressing, but not limited to, the following themes:o Mechanistic insights into the association of pancreatic carcinoma with metabolic dysregulation syndrome components such as obesity-related adipokines, insulin resistance, and lipid disorders, covering oncogenic pathways, oxidative stress, and the tumor microenvironment.o Molecular profiling of diabetes-related pancreatic carcinoma, aiming to identify unique molecular signatures, including gene expression, epigenetic changes, and proteomic characteristics, as potential biomarkers.o Exploring the role of endocrine signaling networks, particularly the insulin-IGF axis and GLP-1 signaling, in pancreatic carcinoma development and therapy responses.o Investigative analyses of the interaction between metabolic dysregulation conditions and tumor metastasis.o Novel therapeutic strategies targeting metabolic and endocrine signaling transduction pathways in pancreatic carcinoma treatment. We invite high-quality original research articles, in-depth reviews, and thought-provoking perspectives centered around these themes.
