Unravelling immune–inflammatory regulation within tissue microenvironments in chronic inflammatory disorders
The field of immunology has long emphasized the critical role of immune and inflammatory pathways in the pathogenesis of chronic inflammatory diseases. These conditions, encompassing autoimmune disorders, chronic musculoskeletal illnesses, and inflammation-driven cancers, are characterized by persistent immune activation and complex molecular signaling within affected tissues. At the heart of chronic inflammation lies a constantly evolving tissue microenvironment, shaped by interactions among immune cells, resident tissue cells, and a network of soluble mediators such as cytokines and chemokines. This dynamic milieu not only drives local tissue remodeling and dysfunction but also perpetuates disease progression. Despite advances in deciphering immune responses at the systemic level, the intricacies of immune and inflammatory dysregulation within specific tissue microenvironments remain incompletely understood. Recent studies highlight striking similarities in immune–inflammatory mechanisms across distinct chronic diseases, suggesting common pathways that could be therapeutically exploited. Yet significant knowledge gaps persist in defining how these localized changes orchestrate chronic inflammation and pathology. This Research Topic seeks to advance our understanding by gathering studies that investigate immune–inflammatory dysregulation within the tissue microenvironment across chronic inflammatory diseases. The central aim is to elucidate how immune cells, local stromal cells, and inflammatory mediators coalesce to sustain inflammation and promote tissue damage, with an emphasis on shared mechanisms that transcend disease-specific contexts. We encourage contributions examining cellular interactions, signaling pathways, and molecular networks that underpin immune-driven changes within bone, joint, spinal, and other tissues prone to persistent inflammation. Additionally, the Research Topic welcomes innovative therapeutic perspectives, especially those targeting common immune–inflammatory pathways or integrating natural products and traditional medicine concepts. By encompassing both fundamental research and translational advances, this initiative aspires to foster new strategies for modulating inflammatory microenvironments and improving clinical outcomes. The scope of this Research Topic is focused on investigating immunological and inflammatory processes specifically within tissue microenvironments associated with chronic inflammatory diseases. Studies should emphasize local, microenvironmental mechanisms rather than systemic aspects alone. To gather further insights within these boundaries, we welcome articles addressing, but not limited to, the following themes:• Mechanisms of immune–inflammatory dysregulation in chronic inflammatory disease• Modulation of tissue microenvironments by immune and inflammatory cues• Immune cell dynamics and cytokine networks in musculoskeletal and other chronically inflamed tissues• Crosstalk between immune and stromal cells driving disease progression and tissue remodeling• Pathways linking chronic inflammation with the development of inflammation-associated cancer• Emerging therapeutic strategies targeting immune–inflammatory microenvironments, including natural products and traditional medicine approaches Topic editors: Xu Yang, Xuekun Fu, Sheng Chen Source: https://www.frontiersin.org/research-topics/77738/unravelling-immune-inflammatory-regulation-within-tissue-microenvironments-in-chronic-inflammatory-disorders
