Gene and Cell Therapy Strategies for Age-Related Diseases
The rapid progression of genetic engineering, regenerative medicine, and pharmacology has positioned gene, cell, and drug-based therapies at the forefront of modern medicine. As populations worldwide live longer, the prevalence of age-related diseases - such as neurodegenerative disorders, cardiovascular decline, metabolic dysfunction, and musculoskeletal deterioration - continues to rise. This Research Topic aims to gather cutting-edge research on advanced genetic, cellular, and pharmacological interventions designed to target, slow, or reverse these debilitating conditions, including combined approaches that reflect current clinical and translational research. To align with the scope of this specialty section, submissions should be grounded in the genetic, genomic, or epigenomic aspects of aging. Traditional therapies often focus on symptom management. This collection seeks to highlight strategies that address the root molecular mechanisms of aging. We welcome studies that utilize gene editing, nucleic acid therapeutics, cell-based models, and pharmacological agents to target primary hallmarks of aging, including genomic instability, telomere attrition, epigenetic alterations, mitochondrial dysfunction, and cellular senescence.By exploring how gene, cell, and drug-based therapies interact with the host genome and epigenome, this Research Topic hopes to bridge the gap between basic molecular biology and translational interventions.Suitable themes include, but are not limited to:- CRISPR-based gene editing and epigenetic editing strategies targeting longevity-associated pathways.- In vivo and ex vivo epigenetic reprogramming and cellular rejuvenation therapies.- Stem cell-based therapies and their effects on host gene expression and tissue regeneration.- Gene therapy approaches targeting DNA damage repair mechanisms and genomic stability.- RNA-based therapeutics (including mRNA, siRNA, and miRNA) regulating metabolic and cellular senescence pathways.- Genetic, cell-based, and pharmacological strategies targeting cellular senescence, including senolytic and senomorphic interventions.- Combined gene-, cell-, and drug-based approaches reflecting current clinical and translational practice.- Viral and non-viral delivery vectors optimized for targeting aging tissues.- Overcoming therapy resistance and improving durability of gene and cell therapies in aged or disease-burdened tissue, including microenvironment-mediated mechanisms of therapeutic failure.- Mitogenomics and therapies directed at nuclear-mitochondrial interactions in aging.- Novel genetic and epigenetic biomarkers used to evaluate the efficacy of therapeutic interventions.- Comparative genomics and machine intelligence approaches to optimize target selection for gene therapies.
