Organoid platforms for testing mitochondrial therapies in aging
Mitochondrial biology and organoid research are increasingly converging to study age-related disease and test potential therapies in human-relevant systems. Mitochondrial dysfunction is associated with altered energy metabolism, cellular stress, and tissue decline, but conventional models may not capture the complexity of human tissues. A further challenge is that cellular reprogramming can erase or attenuate age-associated features in induced pluripotent stem cells (iPSCs), raising questions about how faithfully iPSC-derived organoids model aging. Recent advances in organoid culture and functional profiling offer ways to investigate human tissue biology in three dimensions, but approaches to establish, retain, and validate aging features remain varied. Further work is needed to determine which models and readouts best support robust evaluation of mitochondrial and metabolic interventions. This Research Topic aims to advance the use of organoids to investigate mitochondrial dysfunction in aging and evaluate potential therapeutic strategies. It will bring together research on how aging features can be modeled in iPSC-derived systems, how mitochondrial and cellular stress responses can be measured in 3D tissues, and how organoid results can be assessed against human aging data.To gather further insights in human-relevant organoid models of aging and mitochondrial dysfunction, we welcome articles addressing, but not limited to, the following themes:- Testing mitochondrial and metabolic therapies in organoid models- Strategies to induce or preserve aging features in iPSC-derived organoids- Mitochondrial function and cellular-stress readouts in 3D systems- Functional and molecular profiling of age-related phenotypes in organoids- Comparing organoid findings with human aging datasets, tissues, or clinical evidence- Strengths, limitations, and reproducibility of organoid models for aging research Topic editors: Ankit Kushwaha, Vishnu Agarwal, Amit Kumar Singh Source: https://www.frontiersin.org/research-topics/87230/organoid-platforms-for-testing-mitochondrial-therapies-in-aging
