Smart Polymeric Nanoparticles and Nanogels for Precision Drug Delivery: From Soft-Matter Design to Translational Performance
Recent advances in nanomedicine have brought significant improvements to drug delivery systems, yet persistent hurdles remain, including poor targeting, unpredictable pharmacokinetics, and limited translation to clinical practice. Smart polymer-based soft-matter nanocarriers (such as nanogels, polymersomes, polymeric micelles, and polymer-stabilized liposomes) are emerging as innovative platforms capable of overcoming these challenges. Their tunable structural features, including crosslink density, mesh size, viscoelasticity, and surface hydration or zwitterionicity, enable precise control over drug loading, release mechanisms, and bio-interface interactions. These features empower the systems to respond to biological or external stimuli, evade immune clearance, and traverse complex physiological barriers, placing them at the forefront of future precision medicine strategies. Despite substantial progress in drug delivery, many therapeutic agents remain limited by poor bioavailability, rapid clearance, non-specific distribution, and manufacturing constraints, which undermine clinical efficacy and increase toxicity. Smart polymeric nanocarriers offer a promising solution by integrating tunable structural properties with functional versatility, enabling targeted, controlled, and stimuli-responsive delivery tailored to disease-specific microenvironments. The goal of this Research Topic is to explore how soft-matter design parameters - including polymer architecture, crosslinking, viscoelasticity, and surface chemistry - translate into biological outcomes, such as immune evasion, barrier penetration, biodistribution, and pharmacokinetic/pharmacodynamic (PK/PD) performance. We aim to showcase strategies that bridge materials design with translational endpoints, including manufacturability, stability, safety, and regulatory readiness. By uniting advances from polymer chemistry, nanomedicine, and pharmaceutical engineering, this collection seeks to accelerate the development of next-generation polymeric drug delivery systems that offer improved therapeutic efficacy, minimized side effects, and greater clinical impact. This Research Topic welcomes contributions on the design, synthesis, characterization, and application of smart polymer-based soft-matter nanocarriers for advanced drug delivery. Relevant carrier systems include nanogels, polymersomes, polymeric micelles, and polymer-stabilized liposomes. We particularly invite studies that establish quantitative relationships between material features and biological or translational outcomes. Areas of interest include, but are not limited to:- Quantitative links between material design (crosslinking, mesh size, viscoelasticity, surface hydration) and biological performance- Mechanistic insights into immune evasion, cellular uptake, and transport across biological barriers- Impact of structural parameters on biodistribution, PK/PD profiles, and therapeutic outcome- Translational challenges: manufacturability, scalability, stability, and regulatory considerations- Safety, biocompatibility, and degradability of polymeric nanocarriersWe welcome original research articles, reviews, mini-reviews, perspectives, and methods papers.
